摘 要:贝莱斯芽孢杆菌(Bacillus velezensis)分布广泛、生物活性高,是一种安全可靠的生物防治菌株,降低成本提高细胞生物量是实现Bacillus velezensis广泛应用的关键。采用单因素实验筛选最佳碳源、氮源和无机盐,通过响应面优化Bacillus velezensis G127的高密度发酵条件和工艺。结果表明:最佳培养基组成配方及培养条件为牛肉膏添加量15.2 g/L、糖蜜13.0 g/L、NaCl 5.0 g/L、发酵时间72 h、发酵温度30℃、接种量3.3%、初始pH值5.1,优化后生物量可达5.45×10^(10) CFU/mL,获得的高密度发酵工艺为Bacillus velezensis G127的规模化生产及应用提供保障。Bacillus velezensis is widely distributed and has high biological activity,which is a safe and reliable biocontrol strain.Reducing cost and increasing cell biomass is the key to its wide application.The optimal carbon source,nitrogen source and inorganic salt were selected by single factor experiment,and then the high-density fermentation conditions and process of Bacillus velezensis G127 were optimized by response surface.The optimal medium composition and culture conditions were as follows:beef paste supplemental level 15.2 g/L,molasses 13.0 g/L,sodium chloride 5.0 g/L,fermentation time 72 h,fermentation temperature 30℃,inoculation amount 3.3%,and initial pH value 5.1.The optimized biomass could reach 5.45×10^(10) CFU/mL.The obtained high-density fermentation technology provides guarantee for the large-scale production and application of Bacillus velezensis G127.
