摘 要:目的:筛选出新鲜佛手与干燥佛手挥发性成分的代谢差异物,全面分析佛手中挥发性成分的种类,并探讨评价佛手质量的新路径。方法:采用顶空固相微萃取-气相色谱-质谱联用技术(HS-SPME-GC-MS)对佛手的挥发性代谢物进行定性和相对含量分析。结果:共鉴定出110种与香气有关的挥发性成分,分为10类,其中萜类数量最多,新鲜佛手中萜类占比46.6%,干燥佛手中萜类占比46.8%。通过主成分分析(PCA)方法分析挥发性成分可将新鲜和干燥后的佛手进行区分。正交偏最小二乘法判别分析(OPLS-DA)表明该模型稳定,可用于筛选新鲜、干燥佛手中挥发性成分的代谢差异物。通过建立的OPLS-DA模型,筛选出变量重要性投影值(VIP)大于1的化合物共12种,主要包含了萜类和醇类物质。结论:新鲜佛手和干燥后的佛手挥发性成分存在差异。本研究全面分析了佛手中挥发性成分的种类,指明了佛手挥发性成分的变化规律,可为佛手质量评价及质量标志性成分研究提供依据。Objective:To screen out the metabolic differences of volatile components in fresh and dried Citrus medica var.sarcodactylis,comprehensively analyze the types of volatile components in C.medica var.sarcodactylis,and explore new approaches for assessing its quality.Methods:Headspace solid-phase microextraction gas chromatography-mass spectrometry(HS-SPME-GC-MS)was used for qualitative analysis and determination of relative contents of volatile metabolites.Results:A total of 110 volatile components related to aroma were identified and classified into 10 categories,among which terpenes were the most numerous,accounting for 46.6%in fresh C.medica var.sarcodactylis and 46.8%in dried C.medica var.sarcodactylis.Principal component analysis(PCA)successfully distinguished between fresh and dried C.medica var.sarcodactylis based on their volatile components.Orthogonal partial least squares discriminant analysis(OPLS-DA)indicated that the model was stable and could screen out the differential metabolites of volatile components in fresh and dried C.medica var.sarcodactylis.Through the established OPLS-DA model,a total of 12 compounds with variable importance in projection(VIP)values greater than 1 were screened out,mainly including terpenes and alcohols.Conclusion:There were differences in volatile components between fresh and dried C.medica var.sarcodactylis.This study comprehensively analyzed the types of volatile components in C.medica var.sarcodactylis and indicated the changes in its volatile components,which could provide a basis for the quality evaluation and research on quality marker components of C.medica var.sarcodactylis.
