摘 要:为研究天然抑菌物质对单增李斯特菌(Listeria monocytogenes,Lm)生物膜的抑制作用,本研究首先测定了天然抑菌物质肉桂醛、丁香酚、白藜芦醇和百里醌对Lm ATCC 19115的最小抑菌浓度(minimal inhibitory concentration,MIC)和亚抑菌浓度(sub-inhibitory concentrations,SIC),并通过测定泳动能力、生物膜代谢活性(CCK-8法)和扫描电镜探究了不同浓度(MIC、1/4 MIC和1/8 MIC)下4种天然抑菌物质对该菌株的成膜能力(结晶紫染色法及活菌数法)的影响。结果表明:肉桂醛、丁香酚、白藜芦醇和百里醌对Lm ATCC 19115的MIC分别为:640、1280、400和50μg/mL,Lm ATCC 19115在4种天然抑菌物质中的SICs为1/4 MIC和1/8 MIC;上述4种天然抑菌物质均可抑制Lm ATCC 19115生物膜的形成,且在亚抑菌浓度1/4 MIC下生物膜抑制率仍可达到50%以上;4种天然抑菌物质中,肉桂醛的生物膜抑制效果最佳,其可以通过抑制Lm的泳动能力和生物膜代谢活性有效抑制Lm ATCC 19115生物膜的形成。To investigate the biofilm inhibitory effects of natural antimicrobial substances of Listeria monocytogenes(Lm),our study first determined the minimal inhibitory concentrations(MIC)and sub-inhibitory concentrations(SIC)of the natural antimicrobial substances cinnamaldehyde,eugenol,resveratrol,and thymoquinone against Lm ATCC 19115.The effects of four natural antibacterial substances on the biofilm formation ability of the strain were studied by measuring the swimming ability,biofilm metabolic activity(CCK-8 method)and scanning electron microscopy at different concentrations(MIC,1/4 MIC and 1/8 MIC).The results showed that the MICs of cinnamaldehyde,eugenol,resveratrol,and thymoquinone against Lm ATCC 19115 were 640,1280,400,and 50μg/mL,respectively.The SICs of Lm ATCC 19115 for the four natural antimicrobial substances were 1/4 MIC and 1/8 MIC.All of the above 4 natural antimicrobial substances were found to inhibit the biofilm formation of Lm ATCC 19115,with inhibition rates exceeding 50%even at the sub-inhibitory concentration of 1/4 MIC.Among the four natural antimicrobial substances,cinnamaldehyde exhibited the best biofilm inhibition effect,it effectively inhibited the biofilm formation of Lm ATCC 19115 by inhibiting its swimming motility and biofilm metabolic activity.
