流加培养与氧调控协同提高大肠杆菌产L⁃苏氨酸

摘  要:以实验室1株大肠杆菌为出发菌株,首先将活化后的大肠杆菌菌液按照5%接种量接种至优化后的发酵罐培养基中进行培养,调节控制溶解量不低于15%,并实时流加葡萄糖酸钠、磷酸甜菜碱和玉米浆干粉混合溶液,控制细胞的氧消耗速率(oxygen uptake rate,OUR)为215 mmol/(L·h),通过在线氧消耗速率控制系统、补料控制与OUR调节的作用关系,获得最优流加补料方案:流加25%氨水控制pH值,调整转速和通气控制溶氧15%以上,当溶氧开始回升时流加质量浓度为65%的葡萄糖,起始流加速度1 g(/L·h),控制残糖浓度不超过1.0 g/L。结果表明:L⁃苏氨酸产量提高了35%,达到155 g/L。Escherichia coli as the starting strain was activated and inoculated at an amount of 5%into the optimized medium in the fermenter with the dissolved oxygen no less than 15%.The mixture of sodium gluconate,betaine phosphate,and corn steep liquor powder was fed in real time,and the oxygen uptake rate(OUR)was controlled at 215 mmol/(L·h).According to the relationship of the online OUR control system and the feed control strategies with OUR regulation,the feed-batch scheme was optimized as follows:feeding 25%ammonia for the control of pH,maintaining dissolved oxygen at above 15%by agitation and aeration,feeding 65%glucose at the initial flow acceleration of 1 g/(L·h)when the dissolved oxygen increased,and keeping the residual glucose concentration not exceeding 1.0 g/L.The results showed that the L-threonine yield was increased by 35%,reaching 155 g/L.
