摘 要:目的探讨表没食子儿茶素没食子酸酯(EGCG)对过氧化氢(H2O2)诱导的人脐静脉内皮细胞(HUVEC)氧化应激和凋亡的缓解作用及作用通路。方法体外培养HUVEC,分为对照组、H2O2组(200μmol/L)、H2O2+EGCG(50μmol/L)组、H2O2+EGCG+ML385组(Nrf2特异性抑制剂,2.5μmol/L)。采用CCK-8实验检测细胞活力,活性氧(ROS)染色检测细胞内ROS含量,Hoechst染色试剂盒检测细胞凋亡情况,以及Western blot法检测凋亡相关蛋白Bax和B淋巴细胞瘤-2基因(Bcl2)、核转录因子红系2相关因子2(Nrf2)、血红素加氧酶1(HO-1)、切割型胱天蛋白酶-3(Cleaved Caspase-3)蛋白水平。结果选择50μmol/L EGCG作为后续干预的浓度。与对照组比较,H2O2组ROS水平、细胞凋亡率升高,Nrf2和HO-1蛋白表达水平降低(P<0.05)。与H2O2组比较,H2O2+EGCG组ROS水平、细胞凋亡率及Bax、Cleaved Caspase-3蛋白表达水平降低,Nrf2和HO-1、Bcl2蛋白表达水平升高(P<0.05)。与H2O2+EGCG组比较,H2O2+EGCG+ML385组Nrf2和HO-1以及Bcl2蛋白表达水平降低,Bax和Cleaved Caspase-3蛋白表达水平升高(P<0.05)。结论EGCG能够通过激活Nrf2/HO-1通路延缓H2O2诱导的HUVEC氧化应激及凋亡。Objective To investigate the protective effects and underlying mechanisms of epigallocatechin gallate(EGCG)in alleviating hydrogen peroxide(H2O2)-induced oxidative stress and apoptosis in human umbilical vein endothelial cells(HUVEC).Methods HUVEC were cultured in vitro and divided into four groups:control group,H2O2 group(200μmol/L),H2O2+EGCG group(50μmol/L),and H2O2+EGCG+ML385 group(specific Nrf2 inhibitor,2.5μmol/L).Cell viability was assessed by the CCK-8 assay,intracellular reactive oxygen species(ROS)levels were measured by ROS staining,and apoptosis was detected using the Hoechst staining apoptosis detection kit.Western blot analysis was used to measure the protein levels of apoptosis-related proteins Bax,B-cell lymphoma 2(Bcl2),nuclear factor erythroid 2-related factor 2(Nrf2),heme oxygenase 1(HO-1),and Cleaved Caspase-3.Results A concentration of 50μmol/L EGCG was selected for subsequent interventions.Compared with the control group,the level of ROS and apoptosis rate in the H2O2 group were increased,and the expression levels of Nrf2 and HO-1 proteins were decreased(P<0.05).Compared with the H2O2 group,the ROS level,apoptosis rate and the expression levels of Bax and marker Cleaved Caspase-3 in the H2O2+EGCG group were decreased,and the expression levels of Nrf2,HO-1,and Bcl2 were increased(P<0.05).Compared with H2O2+EGCG group,the expression levels of Nrf2,HO-1,and Bcl2 protein in H2O2+EGCG+ML385 group were decreased,and the expression levels of Bax and Cleaved Caspase-3 protein were increased(P<0.05).Conclusion EGCG can attenuate H2O2-induced oxidative stress and apoptosis in HUVEC by activating the Nrf2/HO-1 pathway.
