阿维菌渣处理工艺优化及其在乳酸乳球菌培养中的应用

摘  要:目的:以阿维菌渣为原料,采用酶-酸复合法对其进行资源化处理,以转化为微生物可利用的营养物质,并评估其作为乳酸乳球菌培养基的可行性。方法:采用酶-酸复合法对阿维菌渣进行降解处理,并通过单因素实验和正交实验优化降解条件。随后,利用降解后的菌渣培养乳酸乳球菌,并采用菌落计数法评估其培养效果。结果:蛋白酶降解蛋白质的最佳工艺条件为菌渣量9 g、加酶量0.10%、水解时间6.0 h;脂肪酶降解脂肪的最佳工艺条件为菌渣量9 g、加酶量0.40%、水解时间4.0 h;酸降解总糖的最佳工艺条件为菌渣量为12 g,加酸量为4.00%,水解时间为3.0 h,温度为110℃。利用降解后的菌渣培养乳酸乳球菌,菌落计数结果为1.2×10^(7)CFU·mL^(-1)。结论:酶-酸复合法可有效降解阿维菌渣,使其转化为可用于乳酸乳球菌培养的营养基质,实现资源化利用,为阿维菌渣的进一步研究和应用提供基础数据。Objective:To utilize avermectin fermentation residue as raw material and treat it by enzyme-acid composite method to transform it into nutrients that can be used by microorganisms,and to evaluate its feasibility as a culture medium for Lactococcus lactis.Method:Avermectin fermentation residue was degraded by enzymeacid composite method,and the degradation conditions were optimized by single factor experiment and orthogonal experiment.Subsequently,Lactococcus lactis was cultured with the degraded residue,and the culture effect was evaluated by colony counting method.Result:The optimal process conditions for protein degradation by protease were 9 g residue,0.10%enzyme amount,and 6.0 h hydrolysis time;the optimal process conditions for fat degradation by lipase were 9 g residue,0.40%enzyme amount,and 4.0 h hydrolysis time;the optimal process conditions for total sugar degradation by acid were 12 g residue,4.00%acid amount,3.0 h hydrolysis time,and temperature of 110℃.The colony count result of Lactococcus lactis cultured with the degraded residue was 1.2×10^(7)CFU·mL^(-1).Conclusion:The enzyme-acid composite method can effectively degrade avermectin fermentation residue and convert it into a nutrient matrix that can be used for the cultivation of Lactococcus lactis,thus realizing resource utilization and providing basic data for further research and application of avermectin fermentation residue.
