鞘内注射辣椒素对SNI大鼠脊髓CGRP的表达影响

摘  要:为了研究鞘内注射辣椒素的神经病理性疼痛大鼠脊髓降钙素基因相关肽(calcitonin gene-related peptide, CGRP)的表达变化及其在疼痛中的作用,将18只大鼠分为正常组(NC group)、SNI大鼠1 d组(SNI 1 d)、SNI大鼠3 d组(SNI 3 d)、SNI大鼠7 d组(SNI 7 d)、SNI大鼠10 d组(SNI 10 d)、SNI大鼠14 d组(SNI 14 d),每组3只,测量大鼠的机械缩足域值(paw withdrawal threshold, PWT),Elisa技术检测脊髓CGRP变化。另将12只大鼠分为正常组、假手术组、鞘内注射DMSO(5μL)组、鞘内注射辣椒素(300 nmol/L)组,观察SNI大鼠PWT变化,脊髓CGRP的表达情况。SNI大鼠PWT显著降低,差异有统计学意义;SNI大鼠脊髓CGRP含量随着时间延长逐渐增加,差异有统计学意义。鞘内注射辣椒素(300 nmol/L)SNI大鼠PWT显著降低,第7 d最低(0.03±0)g,差异有统计学意义;脊髓CGRP含量明显升高[(89.22±9.62) pg/mg],差异有统计学意义。鞘内注射辣椒素(300 nmol/L)增加SNI大鼠脊髓CGRP的表达,表明中枢注射特定剂量辣椒素可促进大鼠的炎症反应,加重疼痛。To investigate the neuropathic pain rat model,the expression and function of spinal cord CGRP(calcitonin gene-related peptide)after intrathecal injection of capsaicin,18 rats were divided into normal group(NC group),SNI rats I d group(SNI1 d),SNI rats 3 d group(SNI3 d),SNI rats 7 d group(SNI 7 d),SNI rats 10 d group(SN 10 d)and SNI rats 14 d group(SNI14 d),with 3 rats in each group.The mechanical foot retraction zone(PWT)was measured,and the CGRP of spinal cord was detected by Elisa.In addition,12 rats were divided into normal group,sham operation group,intravaginal injection of DMSO(5μL)group and intravaginal injection of capsaicin(300 nmol/L,5μL)group.The changes of PWT and the expression of CGRP in the spinal cord of SNI rats were observed.PWT in SNI rats was significantly decreased,the difference was statistically significant,and CGRP content in the spinal cord of SNI rats was gradually increased with the extension of time,the difference was statistically significant.The PWT of rats injected with capsaicin(300 nmol/L)SNI was significantly decreased,the lowest was(0.03±0)g on the 7th day,the difference was statistically significant,and the CGRP content in spinal cord was significantly increased[(89.22±9.62)pg/mL],the difference was statistically significant.Intravaginal injection of capsaicin(300 nmol/L)increased the expression of CGRP in the spinal cord of SNI rats,indicating that central injection of specific dosage of capsaicin can promote the inflammatory response and aggravate pain in rats.
