玉米须抗2型糖尿病网络药理学和药效学评价

摘  要:玉米须(MS)已被证明具有显著的降血糖作用。研究旨在使用网络药理学和分子对接方法探索玉米须治疗T2DM的分子机制,应用动物实验验证玉米须提取物对2型糖尿病大鼠的空腹血糖、胰岛、肝脏病理学改变及PI3K、AKT mRNA表达的影响。TCMSP数据库表明,玉米须包含12种活性成分。PPI和核心靶点分析确定AKT1、TNF、TP53、IL6和PTGS2是玉米须治疗T2DM的关键靶点。采用分子对接方法证实玉米须生物活性分子与T2DM靶点具有高亲和力。富集分析表明:玉米须活性成分参与多种信号通路,主要通路包括脂质和动脉粥样硬化、IL-17信号通路、PI3K-Akt信号通路等。试验证明,玉米须提取物能够控制T2DM大鼠空腹血糖,改善T2DM大鼠胰岛和肝脏组织形态,逆转PI3K和AKT mRNA表达。网络药理学、分子对接以及应用动物实验验证结果表明,玉米须可改善T2DM,其作用机制可能与调节PI3K和AKT mRNA表达有关。Maydis stigma(MS)has been demonstrated to have extremly anti-hyperglycemic effects.The specific multi-target mechanisms through which MS acts in type 2 diabetes mellitus(T2DM)are not yet fully understood.Therefore,this study aimed to explore the molecular mechanism of MS in the treatment of T2DM using network pharmacology and molecular docking methods.Animal experiments verify the effects of MS extract on fasting blood glucose,pancreatic and liver pathological changes,and PI3K and AKT mRNA expression in type 2 diabetic rats.The TCMSP data base indicated that MS contains 12 active components.PPI and core target analysis identified AKT1,TNF,TP53,IL6 and PTGS2 as the key targets of MS in the treatment of T2DM.Furthermore,using the molecular docking method confirmed that the bioactive molecules of MS had a high affinity with the target of T2DM.Enrichment analysis revealed that MS's active components participate in several key signaling pathways,including those related to lipids,atherosclerosis,IL-17,prostate cancer,PI3K-Akt,and bladder cancer.Animal experiments had proven that MS extract could control fasting blood glucose in T2DM rats,improve the morphology of pancreatic and liver tissues in T2DM rats,and reverse the expression of PI3K and AKT mRNA.The results of network pharmacology,molecular docking,and animal experiments verification indicated that MS could improve T2DM,and its mechanism of action might be related to the regulation of PI3K and AKT mRNA expression.
