摘  要:麦角甾醇具有抗炎和抗氧化等多种生理活性。麦角甾醇酯可进一步改善麦角甾醇的熔点、油溶性和稳定性等性能。文章以十二烷基苯磺酸为催化剂,研究了麦角甾醇与月桂酸在有机溶剂正己烷中酯化合成月桂酸麦角甾醇酯的工艺和转化率。通过响应面分析,研究反应温度、反应时间、底物摩尔比(月桂酸:麦角甾醇)和催化剂使用量及其交互作用对月桂酸麦角甾醇酯酯化合成的影响,构建相应的回归模型,并进行验证。结果表明:反应温度79.4℃、反应时间8.5 h、底物摩尔比3.6:1、催化剂用量7.3%为最佳合成条件,验证试验表明转化率可达94.6%。通过红外光谱、薄层色谱和核磁共振分析对结构进行了表征,确认了月桂酸麦角甾醇酯的有效合成。该研究为甾醇酯合成及应用提供了一定的参考依据。Ergosterol possesses various physiological activities,such as anti-inflammatory and antioxidant properties.Ergosterol esters can further improve the properties of ergosterol,such as melting point,oil solubility and stability.In this paper,the process and conversion rate of ergosterol esters of lauric acid were investigated for the synthesis of ergosterol esters of lauric acid by esterification of ergosterol with lauric acid in organic solvent n-hexane using dodecylbenzenesulfonic acid(DBSA)as a catalyst.The effects of reaction temperature,reaction time,substrate molar ratio(lauric acid:ergosterol)and catalyst usage and their interactions on the esterification synthesis of ergosterol esters from lauric acid were investigated by response surface analysis,and the corresponding regression models were constructed and validated.The results showed that the reaction temperature of 79.4℃,the reaction time of 8.5 h,the substrate molar ratio of 3.6:1,and the catalyst dosage of 7.3%were the optimal synthesis conditions.The conversion rate of the validation test could reach 94.6%.Further infrared spectroscopy,thin layer chromatography detection analysis and nuclear magnetic resonance structural characterisation were used to confirm the effective synthesis of ergosterol laurate.This study will provide some references for sterol ester synthesis and its application.
