复合凝聚法制备山奈酚微胶囊及对剧烈运动所致氧化应激损伤保护作用

摘  要:采用复合凝聚法制备海藻酸钠/壳聚糖-山奈酚(Sodium alginate/chitosan-kaempferol,SA/CSKae)微胶囊,利用傅里叶红外光谱、热重分析及稳定性试验考察该微胶囊的性质。通过建立骨骼肌氧化应激损伤小鼠模型,研究该微胶囊的体内抗氧化作用。确定SA/CS-Kae微胶囊的最佳制备条件为海藻酸钠质量浓度3.32%、壳聚糖质量浓度1.62%、氯化钙质量浓度5.34%、海藻酸钠与山奈酚质量比为3.18:1。在此条件下,所得微胶囊的包覆率达到(86.4±0.3)%。傅里叶红外光谱、热重分析及稳定性考察结果显示,山奈酚被包覆在微胶囊内,热稳定性良好,可在室温下长期存放。按照200 mg/kg剂量灌胃,山奈酚能够提高小鼠骨骼肌中超氧化物歧化酶和谷胱甘肽过氧化物酶的活力,有效减少丙二醛的生成量,对骨骼肌氧化应激损伤具有保护作用。与相同灌胃量(200 mg/kg)的山奈酚相比,SA/CS-Kae微胶囊可进一步减轻剧烈运动导致的小鼠骨骼肌氧化应激损伤,其体内抗氧化作用更佳。Sodium alginate/chitosan-kaempferol(SA/CS-Kae)microcapsules were prepared by complex coacervation.The properties of the microcapsules were characterized using Fourier transform infrared spectroscopy(FT-IR),thermogravimetric analysis(TGA),and stability tests.A mouse model of skeletal muscle oxidative stress injury was established to investigate the in vivo antioxidant effects of the microcapsules.The optimal preparation conditions for the SA/CS-Kae microcapsules were as follows:sodium alginate concentration of 3.32%,chitosan concentration of 1.62%,calcium chloride concentration of 5.34%,and a sodium alginate to kaempferol mass ratio of 3.18:1.Under these conditions,the encapsulation efficiency of the microcapsules reached(86.4±0.3)%.Results from FT-IR,TGA,and stability tests demonstrated that kaempferol was successfully encapsulated within the microcapsules,which exhibited good thermal stability and could be stored at room temperature for extended periods.At a gavage dose of 200 mg/kg,kaempferol improved the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the skeletal muscle of mice,effectively reducing malondialdehyde(MDA)levels and providing protective effects against oxidative stress injury in skeletal muscle.Compared with the same gavage dose(200 mg/kg)of kaempferol,SA/CS-Kae microcapsules further alleviated the oxidative stress injury in mouse skeletal muscle caused by strenuous exercise,demonstrating superior in vivo antioxidant activity.
