基于网络药理学分析威麦宁胶囊治疗乳腺癌的作用机制

摘　　要：目的采用网络药理学方法探讨威麦宁胶囊治疗乳腺癌的作用机制。方法通过检索中药系统药理学数据库与分析平台(TCMSP)、查阅文献收集威麦宁胶囊的活性成分,并在TCMSP数据库获得活性成分的潜在靶点;通过GeneCards、OMIM数据库检索乳腺癌疾病靶点;采用Venny平台分析威麦宁胶囊和乳腺癌的共同靶点,将两者的共同靶点通过String数据库进行网络作用分析,将结果导入Cytoscape软件进行可视化分析,构建蛋白质-蛋白质相互作用(PPI)网络以及药物-成分-靶点网络;通过DAVID数据库进行药物疾病共同靶点的基因本体(GO)和京都基因与基因组百科全书(KEGG)通路富集分析。结果获得威麦宁有效成分20个,潜在作用靶点216个;筛选得到威麦宁胶囊抗乳腺癌的核心作用靶点共29个,核心活性成分为槲皮素、没食子酸和木犀草素,主要作用于癌症相关信号通路、HIF1信号通路、PI3K-Akt信号通路等生物学通路。结论威麦宁胶囊通过多成分、多靶点、多通路发挥治疗乳腺癌的作用。Objective To investigate the pharmacological mechanism of Weimaining capsule in the treatment of breast cancer by using a network pharmacology approach.Methods The activecomponents and potential targets of Weimaning capsule were screened out through TCMSP,and supplemented with literature.The gene expression data of BC were obtained from the GeneCards human gene database and online Mendelian Inheritance in man database(OMIM).The common targets were analyzed by Venny platform.A protein-protein interaction(PPI)network of intersection targets was constructed by STRING and visualized by Cytoscape software,and GO functional annotation and KEGG pathway enrichment analysis were performed by database for annotation,visualization and integrated discovery.Results Selecting the OB and DL as filter condition,20 active ingredients and 216 potential targets were involved.Four core components which are quercetin,gallic acid and luteolin and 29 core therapeutic targets of anti-breast cancer were obtained.The KEGG analysis showed that target genes were enriched in several key cancer-related signaling pathways,including the Pathways in cancer,HIF1 signaling pathway,PI3K-Akt signaling pathway,etc.Conclusion The effectiveness of Weimaining capsule in treating breast cancer has multiple components,targets and pathways.
