摘  要:[目的]提高紫球藻胞外多糖(Porphyridium aerugineum exopolysaccharide,EPS)的生理活性及应用范围。[方法]采用浓硫酸法修饰EPS,以硫酸基取代度为指标,利用响应面法对紫球藻胞外多糖硫酸酯(sulfated Porphyridium aerugineum exopolysaccharide,S-EPS)的制备工艺进行优化,同时对S-EPS理化性质进行测定。[结果]最佳工艺条件为反应时间6 h、反应温度26℃、m_(硫酸铵):m_(EPS)=4∶5,此条件下取代度为0.8075±0.0087。红外分析证明EPS被成功硫化。热重分析表明S-EPS的热稳定性优于EPS。粒径与Zeta电位分析表明,取代度越大的样品越稳定,—SO_(3)H基团含量越高,电位绝对值越大,不易聚集。抗氧化活性试验结果显示,S-EPS对DPPH·的清除率为92.17%,明显高于EPS的。[结论]硫化修饰提高了EPS的抗氧化能力,说明硫化修饰是一种较为可行的改善多糖的方法。[Objective]To improve the biological activity and application range of Porphyridium aerugineum exopolysaccharide(EPS).[Methods]EPS was modified using the concentrated sulfuric acid method,with the degree of sulfation as the indicator.The preparation process of sulfated P.aerugineum exopolysaccharide(S-EPS)was optimized using response surface methodology.The physicochemical properties of S-EPS were also determined.[Results]The optimal conditions were a reaction time of 6 h,a reaction temperature of 26℃,and a mass ratio of ammonium sulfate to EPS of 4∶5.Under these conditions,the degree of substitution was 0.8075±0.0087.Infrared analysis confirmed that EPS was successfully sulfated.Thermogravimetric analysis showed that the thermal stability of S-EPS was superior to that of EPS.Particle size and Zeta potential analysis indicated that samples with a higher degree of substitution were more stable,with higher—SO_(3)H group content,larger absolute potential,and less aggregation.The results of the antioxidant activity experiment showed that the DPPH·clearance rate of S-EPS was 92.17%,significantly higher than EPS.[Conclusion]Sulfation modification improved the antioxidant capacity of EPS,indicating that sulfation is a feasible method for enhancing polysaccharides.
