酿酒葡萄皮渣提取物对癌细胞的抑制研究

摘  要:为探究酿酒葡萄皮渣提取物对人体癌细胞的抑制作用,以甲醇为溶剂对酿酒葡萄皮渣进行提取,使用MTT比色分析法、DAPI染色法、AnnexinV-FITC/PI双染法分析皮渣提取液对癌细胞存活率、细胞核形态、细胞周期和细胞凋亡方面的影响。结果表明,葡萄皮渣提取物对于不同的癌细胞作用不一,对A549(人非小细胞肺癌细胞株)细胞的抑制作用最明显;对A549细胞进一步研究发现,10、15μg·mL^(-1)浓度的提取液导致A549细胞数量明显减少,细胞核皱缩;A549细胞的S期缩短,G1期和G2期延长。综合分析可知,葡萄皮渣提取物导致A549细胞形态异常,可能是其抑制A549细胞的机制之一。To study the inhibitory effect of wine grape peel extract on human cancer cells,wine grape peel residue was extracted using methanol as solvent.MTT colorimetric analysis,DAPI staining and Annexin V-FITC/PI double staining were used to analyze its effects on cancer cell survival rate,cell morphology,cell cycle and apoptosis.The results showed that grape peel extract had different effects on different cancer cells,with the most significant inhibitory effect on A549 cells.Further study of A549 cells found,10、15μg·mL^(-1) of the extract concentration resulted in a significant decrease in the number of A549 cells and nuclear shrinkage;The S phase of A549 cells was shortened,while the G1 and G2 phase were lengthened.Comprehensive analysis showed that grape peel extract caused abnormal morphology of A549 cells,which may be one of the mechanisms of inhibiting A549 cells.
