摘  要:类胡萝卜素作为一类天然色素具有广泛的应用,而红酵母发酵生产类胡萝卜素具有独特的优势。以海洋源胶红酵母H118为研究对象,采用Plackett-Burman实验设计对影响类胡萝卜素产生的9个因素进行统计分析,影响极显著的因素为培养温度,对其进行梯度优化后确定为25℃。在此基础上,采用优化的工艺条件经5 L发酵罐批培养,发酵至36 h和48 h的类胡萝卜素质量浓度分别达1591.11,1627.92μg/L,该结果可为后续类胡萝卜素产业化生产及其应用提供技术支撑。Carotenoids are widely used as natural pigments,and Rhodotorula has unique advantages for producing carotenoids through fermentation.The marine Rhodotorula mucilaginosa H118 was used as the research subject,and the nine factors affecting the production of carotenoids were statistically analyzed with the Plackett-Burman experimental design,and the culture temperature was identified as the most significant factor and it was optimized to 25℃through gradient optimization.Based on this,the optimized process conditions of Rhodotorula mucilaginosa H118 were adopted for batch culture in a 5 L fermenter,and the carotenoid content reached 1591.11μg/L and1627.92μg/L with fermentation for 36 h and 48 h,respectively,providing technical support for the subsequent large-scale production and application of carotenoids.
