摘 要:Tight junctions are crucial intercellular connections between epithelial and certain endothelial cells that play a vital role in maintaining cell polarity,regulating paracellular permeability,and preventing pathogens from crossing the epithelial and endothelial layers[1],[2],[3].Tight junction dysfunction is associated with various diseases,including inflammatory bowel disease,tumor metastasis,and neurodegenerative diseases[4].Therefore,a deeper understanding of the regulatory mechanisms of tight junctions is important for disease prevention and treatment.Researchers have identified key proteins necessary for the formation and positioning of tight junctions using genetic and biochemical approaches;these include adhesion receptors from the claudin family and two scaffold proteins,zonula occludens proteins ZO1 and ZO2[5],[6].Previous studies have shown that the phase separation of ZO proteins into a condensed,membrane-bound phase is essential for claudin receptor organization and tight junction formation[7],[8].However,tight junctions must maintain stability under diverse physiological and pathological conditions while also being capable of swiftly responding to environmental shifts.How they achieve this balance between stability and plasticity,and the regulatory mechanisms underlying this equilibrium,pose significant scientific questions.
