摘  要:为开发利用高粱资源,探究超声波辅助-酶解法对高粱总黄酮提取率的影响,采用响应面法优化了提取工艺,并评价其抗氧化抑菌活性。通过单因素实验结果,利用Box-Behnken方法进行三因素三水平响应面实验,得到最佳工艺条件为乙醇浓度67.0%,提取温度50.6℃,提取时间51.9 min,此时高粱总黄酮提取率为1.2%。高粱黄酮提取物抗氧化性活性结果表明,总还原力与黄酮浓度呈正相关关系,当黄酮提取物浓度为0.02 g/L时,对DPPH·的清除率达到了90%以上,·OH的清除率达到了60%以上。高粱黄酮提取物对金黄色葡萄球菌的抑制作用最大,对大肠杆菌的抑制作用次之,而对单增李斯特菌则无抑制作用。该结果可为高粱提取物作为天然抗氧化活性物质的应用研究提供参考,为新型饲料添加剂的开发提供思路。To explore the effect of ultrasound assisted enzymatic hydrolysis on the extraction rate of total flavonoids from sorghum resources,response surface methodology is used to optimize the extraction process and evaluate its antioxidant and antibacterial activities.Using a single factor experiment result,a three factor three-level response surface test is conducted by the Box-Behnken method,and the optimal process conditions are obtained as ethanol concentration of 67.0%,extraction temperature of 50.6℃,and extraction time of 51.9 minutes.The total flavonoid extraction rate of sorghum is 1.2%.The antioxidant activity of sorghum flavonoid extract shows a positive correlation between total reducing power and flavonoid concentration.When the concentration of flavonoid extract is 0.02 g/L,the scavenging rate of DPPH·reaches over 90%,and the scavenging rate of·OH reaches over 60%.Sorghum flavonoids extract has the greatest inhibitory effect on Staphylococcus aureus,follows by Escherichia coli,and has no inhibitory effect on Listeria monocytogenes.This result can provide reference for the application research of sorghum extract as a natural antioxidant active substance,and provide ideas for the development of new feed additives.
