摘 要:[目的]探讨丙酮酸激酶M2(pyruvate kinase M2,PKM2)和PD-L1在前列腺癌组织中的表达及其相关性。[方法]采用免疫组化(immunohistochemistry,IHC)技术,对58例前列腺癌患者肿瘤组织及58例良性前列腺增生组织切片进行PKM2和PD-L1的表达检测。根据染色强度和阳性细胞百分比,量化两种蛋白的表达水平。分析PKM2和PD-L1表达与临床病理特征(包括TNM分期、年龄、前列腺特异性抗原、淋巴结转移和Gleason评分)之间的关系。采用SPSS 26.0统计软件进行数据分析。[结果]PKM2和PD-L1在前列腺癌组织中的阳性表达率显著高于良性前列腺增生组织(PKM2:89.7%vs 15.5%,P<0.001;PD-L1:79.3%vs 41.4%,P<0.001)。前列腺癌组织中PKM2的表达与Gleason评分和TNM分期有关(P<0.001,P=0.034),PD-L1的表达也与Gleason评分和TNM分期有关(P<0.001,P=0.035);两者的表达均与患者年龄、前列腺特异性抗原水平和淋巴结转移无关(P均>0.05)。前列腺癌组织中PD-L1与PKM2表达呈正相关(r=0.8141,P<0.001)。[结论]PKM2和PD-L1在前列腺癌中的高表达与肿瘤进展相关。PKM2可能通过增加PD-L1的表达而参与免疫调控。[Objective]To investigate the expression of pyruvate kinase M2(PKM2)and programmed death-ligand 1(PD-L1)in prostate cancer(PCa)and benign prostatic hyperplasia(BPH)tissues,and to explore their clinicopathological significance.[Methods]The expression of PKM2 and PD-L1 in tissue samples from 58 PCa patients and 58 BPH patients were detected by immunohistochemistry(IHC).The relationship between the expression of PKM2 and PD-L1 and clinicapathological features,including TNM stage,prostate-specific antigen(PSA),lymph node metastasis and Gleason score were analyzed.Statistical analysis was carried out by SPSS 26.0 statistical software.[Results]The positive expression rates of PKM2 and PD-L1 in PCa tissue were significantly higher than those in BPH tissue(PKM2:89.7%vs 15.5%,P<0.001;PD-L1:79.3%vs 41.4%,P<0.001).The expression of PKM2 in PCa was correlated to Gleason score and TNM stage(P<0.001,P=0.034),and the expression of PD-L1 was also correlated to Gleason score and TNM stage(P<0.001,P=0.035).While the the expression of PKM2 and PD-L1 was not correlated with the patient age,PSA level and lymph node metastasis(all P>0.05).PD-L1was positively correlated with PKM2 expression in PCa tissue(r=0.8141,P<0.001).[Conclusion]The expression of PKM2 and PD-L1 is associated with the progression of PCa.PKM2 may be involved in immune regulation by enhancing the expression of PD-L1.
