碱性玉米皮可溶性多糖提取及酶解还原糖的测定

摘  要:为更好地开发利用玉米资源,采用膜分离法提取了阿魏酸后的玉米皮渣为试验材料,通过对比分析来考察不同浸提时间、浸提温度对玉米皮多糖提取率的影响。结果表明:最佳提取工艺条件为温度100℃,浸提时间60 min,在这个条件下多糖提取率达6.55%。并用质量分数为0.5%的木聚糖酶酶解剩下的玉米皮渣,使用DNS比色法分析在不同酶解时间条件下还原糖的浓度。试验中,酶解8 h时还原糖浓度最高,其质量分数为170.10 mg/g。In order to better exploit and utilize maize resources,the membrane separation method was used to extract ferulic acid from maize husk residue.The effects of different extraction time and temperature on the extraction rate of maize husk polysaccharide were investigated.The results showed that the optimal extraction conditions were as follows:temperature 100℃,extraction time 60 min,under this condition,the extraction yield of polysaccharide reached 6.55%.The remaining maize husk residue was enzymolized with 0.5%xylanase,and the concentration of reducing sugar under different enzymolysis time was analyzed by DNS colorimetry.In the experiment,the concentration of reducing sugar was the highest at 8 h,and its mass fraction was 170.10 mg/g.
