摘 要:目的探讨养胃颗粒治疗慢性萎缩性胃炎(CAG)的潜在活性成分和作用机制。方法通过中药系统药理学分析平台(TCMSP)检索养胃颗粒的潜在活性成分和作用靶点,通过GeneCards、TTD数据库获取CAG相关疾病靶点,将共同靶点与潜在活性成分导入Cytoscape 3.10.1软件构建中药-成分-共同靶点-CAG相互作用网络,通过String 11.0数据库获得共同靶点蛋白相互作用网络及核心靶点;通过David数据库进行基因本体论(GO)功能富集和京都基因与基因组百科全书(KEGG)通路富集分析。运用Autodock vina软件对度值排名前五的潜在活性成分及靶点进行分子对接验证。结果筛选获得养胃颗粒137个潜在活性成分,CAG与2765个靶点相关,潜在活性成分与CAG有170个共同靶点。核心成分包括槲皮素、木犀草素、山柰酚等,核心靶点包括IL-6、TNF、TP53等。KEGG富集分析主要涉及癌症的发病途径、AGE-RAGE信号通路在糖尿病并发症中的作用等。分子对接结果显示,度值前五的成分与靶点具有较好的结合活性。结论养胃颗粒治疗CAG的主要机制可能通过槲皮素等核心成分作用于IL-6等靶点发挥作用,为深入阐明养胃颗粒的药效物质基础及作用机制提供参考。Objective To investigate the potential active ingredients and mechanism of Yangwei granules in treating chronic atrophic gastritis(CAG).Methods The potential active components and targets of Yangwei granules were searched by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and CAG related disease targets were obtained by GeneCards and TTD databases.The common target and potential active ingredients were imported into Cytoscape 3.10.1 software to construct the TCM-component-common target-CAG interaction network,and the common target protein interaction network and core targets were obtained through String 11.0 database.Genetic ontological(GO)functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed using the David database.Autodock vina software was used to verify the molecular docking of the top five potential active components and targets.Results 137 potential active components of Yangwei granules were selected.CAG was associated with 2765 targets,and there were 170 common targets between CAG and CAG.Core components included quercetin,luteolin,kaempferol,etc.Core targets included IL-6,TNF,TP53,etc.KEGG enrichment analysis mainly involved the pathogenesis of cancer and the role of AGE-RAGE signaling pathway in diabetic complications.The results of molecular docking showed that the top five components had better binding activity with the target.Conclusion The main mechanism of Yangwei granules in the treatment of CAG may be through the action of quercetin and other core components on IL-6 and other targets,which provides a reference for further elucidating the pharmacodynamic material basis and mechanism of Yangwei granules.
