摘 要:文章建立高效液相色谱法对不同乳源中α-乳白蛋白(α-La)和β-乳球蛋白(β-Lg)进行定性、定量检测。以0.15%三氟乙酸水溶液和0.15%三氟乙酸乙腈溶液对2种蛋白进行梯度洗脱,柱温为35℃,对目标蛋白进行分离检测,以出峰面积进行定量分析。结果表明,2种蛋白线性关系良好(R^(2)>0.99),经优化后的方法蛋白色谱图峰形明显,样品分离程度高,蛋白回收率为92.8%~99.3%,α-La和β-Lg检出限分别为0.0933 mg/mL和0.0765 mg/mL,定量限分别为0.0787 mg/mL和0.0821 mg/mL。该方法分析速度快,灵敏度高,能准确测定不同乳源中α-La和β-Lg含量。试验检测了不同乳中的α-La和β-Lg,其中生牛乳中α-La和β-Lg含量分别为0.143 mg/mL和0.421 mg/mL,均高于灭菌乳;骆驼乳中α-La含量为0.192 mg/mL,高于牛乳和羊乳;β-Lg含量为0.017 mg/mL,低于牛乳和羊乳。A high performance liquid chromatographic(HPLC)method was developed for the qualitative and quantitative determination ofα-lactalbumin(α-La)andβ-lactoglobulin(β-Lg)in different milk sources.The gradient elution of the two proteins was carried out with 0.15%aqueous trifluoroacetic acid and 0.15%acetonitrile trifluoroacetic acid,and the column temperature was 35℃.The target proteins were separated and detected,and the quantitative analysis was performed by the peak area.The results showed that the linear relationship between the two proteins was good(R^(2)>0.99),and the protein chromatograms of the optimized method showed obvious peak shapes and high degree of sample separation,with protein recoveries ranging from 92.8%to 99.3%,and the limits of detection ofα-La andβ-Lg were 0.0933 mg/mL and 0.0765 mg/mL,respectively,and the limits of quantification were 0.0787 mg/mL and 0.0821 mg/mL,respectively.The method is fast and sensitive,and can accurately determine the contents ofα-La andβ-Lg in different milk sources.Theα-La andβ-Lg in different milks were detected in the experiment,and the contents ofα-La andβ-Lg in raw cow's milk were 0.143 mg/mL and 0.421 mg/mL,which were higher than that in sterilized milk;the content ofα-La in camel's milk was 0.192 mg/mL,which was higher than that in cow's milk and goat's milk;and the content ofβ-Lg was 0.017 mg/mL,which was lower than that in cow's milk and goat's milk.
