摘  要:为筛选高产菌体蛋白的菌株并确定其发酵生产单细胞蛋白的最佳条件,本试验从发酵食品残渣中进行筛选,经形态学及ITS序列分析进行菌株鉴定。以不同碳源、氮源、发酵pH、发酵温度及中和补剂进行单因素试验,在此基础上,选择碳、氮源添加量、种子液培养时间、接种量、发酵时间、搅拌速度,进行发酵条件优化,并对最佳条件下生产的单细胞蛋白进行检测。结果表明:筛选到一株能利用廉价非蛋白氮的高产单细胞蛋白菌株Z2,确定为尖孢镰刀菌(Fusarium oxysporum)。在以6.6%的蔗糖为碳源,2%的硫酸铵为氮源,流加20%氨水,接种量11%,搅拌速度为200 r/min,30℃发酵75 h时菌体干重最高,为10.86 g/L,蛋白产量为57.6%,生产的单细胞蛋白无毒素且营养丰富。本研究为单细胞蛋白生产提供一种新的选择,确定最优发酵条件,为提高镰刀菌属发酵生物量及蛋白产量提供技术支持。In order to screen fungi with high protein production and explore the optimum fermentation conditions for single cell protein,the strains were screened from fermented food waste and identified by morphological observation and ITS sequence analysis.Single factor experiments were carried out under different carbon source,nitrogen source,pH,temperature and neutralization supplement conditions.On this basis,carbon or nitrogen source addition amount,inoculate age,inoculate size,fermentation time and shake rate were selected to optimize fermentation conditions,and the single cell protein produced under the optimal conditions were detected.The results showed that the Z2 strain was identified as Fusarium oxysporum.The optimum result of protein production obtained under conditions as follow:6.6%sucrose as carbon source,2%ammonium sulfate as nitrogen source,20%ammonia solution,11%inoculum,shake rate 200 r/min and 30℃for 75 h.The cell dry weight of Z2 reached 10.86 g/L,the protein yield reached 57.6%,and the single cell protein was free of toxin and rich in nutrients.This study provided a new choice for single cell protein production and technical support for increasing Fusarium fermentation biomass and protein yield.
