摘 要:本试验从关中黑猪粪便中分离出192株菌株,通过高通量显微形态观察和分子生物学鉴定筛选得到8株戊糖片球菌(P.pentosaceus)ST98、ST110、ST119、ST124、ST128、ST132、ST135、ST143。8株菌株均具有良好的生长性能、产酸能力和生物被膜形成能力。在pH 3.0的环境中,菌株ST135的存活率大于100%,其余菌株的存活率均达60%以上;在0.20%的胆盐浓度下,菌株ST124的存活率达79.41%,其余菌株的存活率大于100%;0.30%的胆盐浓度下,菌株ST98、ST135的存活率大于100%,菌株ST124、ST132、ST143的存活率大于70%;体外抑菌试验结果显示,8株菌均能抑制沙门氏菌和金黄色葡萄球菌的生长,其中ST124对金黄色葡萄球菌的抑制能力最强,抑菌圈可达15 mm。研究结果为进一步研究戊糖片球菌的功能特性和生产应用奠定了科学基础,为猪用益生菌提供了候选菌株资源。In this experiment,192 strains were isolated from the feces of Guanzhong black pigs.Eight strains of Pediococcus pentosaceus,namely ST98,ST110,ST119,ST124,ST128,ST132,ST135 and ST143 were identified through high throughput microscopic morphological observation and molecular biological identification.Further research revealed that these eight strains exhibited promising growth performance,acid production ability and biofilm formation ability.What’s more,the survival rate of ST135 exceeded 100%under pH 3.0 conditions while the other strains had a survival rate of more than 60%.Meanwhile,when exposed to a bile salt concentration of 0.20%,strain ST124 achieved a survival rate of 79.41%while the other seven strains had a survival rate greater than 100%.At a bile salt concentration of 0.30%,the survival rate of ST98 and ST135 was greater than 100%,and the survival rate of ST124,ST132 and ST143 was greater than 70%.The in vitro antibacterial tests demonstrated that all eight strains of Pediococcus pentosaceus inhibited the growth of both Staphylococcus aureus and Salmonella.Among them,ST124 displayed the most potent inhibitory ability against Staphylococcus aureus,with an inhibitory zone measuring up to 15 mm.The findings established a scientific groundwork for further investigation into the functional attributes and production application of Pediococcus pentosaceus.Moreover,it provided the candidate strain resources for probiotics of swine.
