摘 要:为明确猕猴桃皮多酚提取物(kiwifruit peel polyphenols extract,KPE)对脂多糖应激大鼠肠道屏障功能的影响及其作用机制,将24只大鼠分为对照组、脂多糖(lipopolysaccharide,LPS)组和猕猴桃皮多酚干预(KPE-LPS)组,进行4周饲喂实验,实验结果显示,与对照组相比,LPS损伤组结肠组织大量炎性细胞浸润,杯状细胞数量减少,且血清二胺氧化酶(diamineoxidase,DAO)含量升高51.48%,黏蛋白MUC1表达含量显著降低。经KPE干预后,肠道组织结构清晰,杯状细胞数量增多,DAO含量下降至7.54 U/L,黏蛋白MUC1平均光密度值和黏蛋白MUC1 mRNA表达含量升高;同时,经KPE干预后,肠道氨氮含量显著降低,乙酸、丙酸、丁酸含量分别上升至2.02、0.51、0.64 mg/g。16S rRNA高通量测序结果显示,猕猴桃皮多酚干预对肠道菌群构成有重塑作用,能促进有益菌异杆菌属和经黏液真杆菌属等的生长,抑制有害菌瘤胃球菌科和毛螺菌科等的增殖。综上,猕猴桃皮多酚可以通过改善肠道屏障功能和调节肠道菌群构成来缓解脂多糖应激造成的肠道健康问题。This study aimed to clarify the effect of kiwifruit peel polyphenol extract(KPE)on intestinal barrier function and its mechanism of action in lipopolysaccharide(LPS)induced stressed rats.Twenty-four rats were divided into the control group,the LPS damage group,and the KPE intervention group(KPE-LPS)for a 4-week feeding experiment.The experimental results showed that,compared with the control group,the LPS damage group exhibited significant infiltration of inflammatory cells in the colonic tissues,a reduction in the number of goblet cells,a 51.48%increase in serum diamine oxidase(DAO)content,and a significant decrease in the expression of mucin MUC1.After KPE intervention,the intestinal tissue structure became clearer,the number of goblet cells increased,DAO content decreased to 7.54 U/L,and the average optical density expression and mRNA expression levels of mucin MUC1 were elevated.Additionally,KPE intervention significantly reduced the intestinal ammonia nitrogen content,and the content of acetic acid,propionic acid,and butyric acid increased to 2.02,0.51,and 0.64 mg/g,respectively.The results of 16S rRNA high-throughput sequencing revealed that KPE intervention had a remodeling effect on the intestinal flora composition,promoting the growth of beneficial bacteria,such as Allobaculum and Blautia,while inhibiting the proliferation of harmful bacteria,such as Ruminococcaceae and Lachnospiraceae.KPE can alleviate intestinal health issues caused by LPS stress by improving intestinal barrier function and modulating intestinal flora composition.
