摘 要:天然、安全、高效的抗氧化剂对于防止氧化应激具有重要价值。基于此,本文采用固相合成法制备了一种新型抗氧化肽GINYWLAHK,分子量为1101.26 Da。通过对ABTS阳离子自由基清除能力、H2O2诱导的HepG2细胞保护作用及细胞内活性氧(Reactive Oxygen Species,ROS)水平和分子对接技术来评价GINYWLAHK的抗氧化活性。结果表明,抗氧化肽GINYWLAHK在2 mg·mL^(-1)时,ABTS阳离子自由基清除率为92.92%±0.12%,同时,抗氧化肽GINYWLAHK还能抑制H2O2诱导HepG2细胞的氧化损伤,显著减少活性氧积累;分子对接结果显示抗氧化肽GINYWLAHK可与Keap1活性口袋中的关键氨基酸残基相互作用,具备调节体内抗氧化作用的潜力。该研究可为抗氧化肽作为新型抗氧化剂应用于功能食品提供理论依据。Natural,safe and highly effective antioxidants are of great value in preventing oxidative stress.Based on this,a novel antioxidant peptide GINYWLAHK with a molecular weight of 1101.26 Da was prepared by solid-phase synthesis.The antioxidant activity of GINYWLAHK was comprehensively evaluated from multiple aspects.These included its free radical scavenging ability against ABTS,the protective effect it exerted on H2O2-induced HepG2 cells,the impact on the intracellular reactive oxygen species(ROS)level,and by means of the molecular docking technique.The results showed that the free radical scavenging rate of ABTS reached 92.92%±0.12%at 2 mg·mL^(-1).Meanwhile,GINYWLAHK could inhibit oxidative damage on H2O2-induced HepG2 cells and significantly reduce the accumulation of reactive oxygen species.Molecular docking results showed that GINYWLAHK can interact with key amino acid residues in the active pocket of Keap1,which has the potential to regulate the antioxidant effect in vivo.This study can provide theoretical basis for the application of antioxidant peptides as new antioxidants in functional foods.
