摘 要:试验旨在研究饲粮铁含量对乌金猪内脏铁沉积和肠道屏障功能的影响。选取18头35日龄、体重相近且健康的乌金猪仔猪,随机分为3组,每组6个重复,每个重复1头仔猪。A组饲喂基础饲粮,B组饲喂基础饲粮+100 mg/kg甘氨酸铁(以铁计),C组饲喂基础饲粮+200 mg/kg甘氨酸铁。预试期5 d,正式试验期30 d。结果显示:C组乌金猪十二指肠和盲肠铁含量显著高于A组(P<0.05);B组十二指肠铁含量显著低于C组(P<0.05)。C组十二指肠绒毛高度显著高于A组和B组(P<0.05);与B组相比,C组隐窝深度显著升高(P<0.05),A组十二指肠隐窝深度显著降低(P<0.05)。与C组相比,A组十二指肠闭锁小带蛋白-1(ZO-1)、闭合蛋白-5(Claudin-5)、白细胞介素-10(IL-10)、干扰素-γ(IFNG)和谷胱甘肽过氧化酶1(GPX1)mRNA相对表达量显著降低(P<0.05);B组十二指肠ZO-1、IFNG、肿瘤坏死因子-α(TNF-α)、黏蛋白2(MUC-2)和GPX1 mRNA的相对表达量显著降低(P<0.05);A组十二指肠IFNG mRNA的相对表达量显著低于B组(P<0.05)。研究表明,饲粮中添加200 mg/kg的甘氨酸铁可以提高乌金猪仔猪十二指肠和盲肠的铁沉积量,改善十二指肠绒毛高度和隐窝深度,上调肠道屏障相关基因表达水平,提高肠道免疫屏障功能。The experiment was designed to investigate the effects of dietary iron content on visceral iron deposition and intestinal barrier function in Wujin pigs.Eighteen 35-day-old Wujin piglets of similar body weight and in good health were selected and randomly divided into three groups,with six replicates per group and one piglets per replicate.The piglets in Group A were fed a basal diet,the piglets in Group B were fed a basal diet+100 mg/kg ferrous glycinate(as iron)and the piglets in Group C were fed a basal diet+200 mg/kg ferrous glycinate.The pre-test lasted for five days and the formal test lasted for 30 days.The results showed that the iron content in the duodenum and cecum of Wujin pigs in Group C was significantly higher than that in Group A(P<0.05).The duodenal iron content in Group B was significantly lower than that in Group C(P<0.05).The villus height in the duodenum of Group C was significantly higher than that in Groups A and B(P<0.05).Compared with Group B,the crypt depth in Group C was significantly increased(P<0.05),while the duodenal crypt depth in Group A was significantly decreased(P<0.05).Compared with Group C,the relative mRNA expression levels of atresia zonulin-1(ZO-1),closure protein-5(Claudin-5),interleukin-10(IL-10),interferon-γ(IFNG),and glutathione peroxidase 1(GPX1)in the duodenum of Group A were significantly reduced(P<0.05).The relative mRNA expression levels of ZO-1,IFNG,tumor necrosis factor-α(TNF-α),mucin 2(MUC-2),and GPX1 in the duodenum of Group B were significantly reduced(P<0.05).The relative mRNA expression level of IFNG in the duodenum of Group A was significantly lower than that in Group B(P<0.05).The study indicates that the addition of 200 mg/kg ferrous glycinate to the diet can increase iron deposition in the duodenum and cecum,enhance duodenal villus height and crypt depth,upregulate the expression levels of intestinal barrier-related genes,and improve intestinal immunity barrier function of Wujin piglets.
