摘 要:建立了高效阴离子交换色谱-脉冲安培法测定食品中15种功能糖及糖醇的分析方法。样品经超声提取、沉淀蛋白后,采用CarboPac PA10(4×250mm)色谱柱分离,在流速为0.90 mL·min^(-1),柱温为25℃的梯度洗脱条件下进行分析,外标法定量。15种功能糖及糖醇在0.25 mg·L^(-1)~50.0 mg·L^(-1)浓度范围内线性相关系数良好(r≥0.999),方法的定量限为0.08 mg/100g~1.74 mg/100g。以饮料、饼干、巧克力和奶粉为样品基质,在3个不同加标水平下,15种功能糖及糖醇的回收率为90.1%~109.3%,相对标准偏差为0.27%~3.80%。该方法在20 min内即可实现15种功能糖及糖醇的定量分析,操作简便、分析速度快,灵敏度高,可以应用于食品中多种功能糖及糖醇含量的测定。A method for the determination of 15 functional sugars and sugar alcohols in food by high performance anion exchange chromatography with pulsed amperometry was established.After ultrasonic extraction and protein precipitation,the samples were separated by CarboPac PA10(4×250mm)column,and were analyzed under the gradient elution condition of 0.90 mL·min^(-1)flow rate and 25℃column temperature.The 15 functional sugars and sugar alcohols were quantified based on external standard method.The experimental results demonstrated good linearity for 15 functional sugars and sugar alcohols in the concentration range of 0.25~50.0 mg·L^(-1),and the correlation coefficients(r)were above 0.999.The quanification limits of 15 functional sugars and sugar alcohols were 0.08 mg/100g~1.74 mg/100g.The average recoveries of 15 functional sugars and sugar alcohols in blank samples(beverages,biscuits,chocolate,and milk powder)were 90.1%~109.3%,and the relative standard deviations were 0.27%~3.80%under three different spike levels.The separation of 15 functional sugars and sugar alcohols could be achieved within 20 minutes,meanwhile the method was simplicity,quickly,and high sensitivity,and successfully applied for determination of functional sugars and sugar alcohols in food.
