摘  要:目的:建立高效液相色谱法检测鸡精中7种鲜味剂的方法。方法:采用高效液相色谱法,用反相C18柱色谱柱分离,以甲醇-磷酸二氢钠水溶液为流动相,梯度洗脱二极管阵列检测器检测。测定鸡精中的7种鲜味剂的含量。结果:待测样液中鲜味剂的响应值在标准曲线范围内,在优化后的液相色谱条件下,7种鲜味剂分离效果良好,鸡精样品中检测出鸟苷酸(Guanosine-5’-monophosphate Disodium Salt,GMP)和肌苷酸(Inosine Monophosphate,IMP)。IMP测定的相对标准偏差在0.78%~1.25%,平均回收率在99.97%~100.00%;GMP测定的相对标准偏差在1.05%~3.01%,平均回收率在99.85%~100.22%。准确度、精密度均满足分析要求,分析结果准确度及精密度高。结论:本次研究有助于更好地指导鸡精的生产和产品质量的控制,具有一定的研究价值。Objective:To establish a HPLC method for the determination of 7 kinds of umami agents in chicken essence.Method:The separation was carried out by high performance liquid chromatography(HPLC)with reversed phase C18 column.The aqueous solution of methanol-sodium dihydrogen phosphate was used as mobile phase,and the detection was carried out by gradient elution diode array detector.The content of 7 kinds of umami agents in chicken essence was determined,and the existence of other nucleotides was also determined.Result:The response values of the umami agents in the samples fell within the range of the standard curve.Under optimized liquid chromatography conditions,the seven umami agents were well separated.Specifically,Guanosine-5’-Monophosphate Disodium Salt(GMP)and Inosine Monophosphate(IMP)were detected in the chicken essence samples.The relative standard deviation(RSD)for IMP was between 0.78%and 1.25%,with an average recovery rate of 99.97%to 100.00%.For GMP,the RSD ranged from 1.05%to 3.01%,and the average recovery rate was between 99.85%and 100.22%.These results indicate that the method’s accuracy and precision meet the required analytical standards.Conclusion:This study is helpful to better guide the production of chicken essence and product quality control,and has certain research value.
