摘  要:为探究罗汉果甜苷(MGE)对湖羊肌肉能量代谢酶活性、肌纤维类型构成以及脂肪代谢相关基因表达的影响,试验选取45只6月龄健康湖羊,分为CON组(基础饲料)、MGE900组(添加900 mg/kg MGE)和MGE1200组(添加1200 mg/kg MGE),共56 d。试验结束后,屠宰取湖羊皮下脂肪、背最长肌和肝脏。测定背最长肌中的琥珀酸脱氢酶(SDH)、乳酸脱氢酶(LDH)和苹果酸脱氢酶(MDH)活性,并检测肌纤维类型组成(MYHCⅠ、MYHCⅡa、MYHCⅡb和MYHCⅡx),肌纤维类型转化相关基因PGC-1α,皮下脂肪、背最长肌、肝脏中可合成脂肪的PPARγ、ACC、FAS基因及分解脂肪ATGL、HSL、LPL基因的表达水平。结果显示:MGE组MYHCI、MYHCⅡa、MYHCⅡx和PGC-1α基因表达增加,SDH活性上升,而MYHCⅡb基因表达和LDH活性降低。MGE组中PPARγ、ACC、FAS基因在背最长肌中表达显著增加,而PPARγ、ACC基因在皮下脂肪和肝脏中表达降低。ATGL、HSL、LPL基因在MGE组背最长肌表达下降,但在皮下脂肪和肝脏中上升。研究结果表明,罗汉果甜苷可调控湖羊肌肉基因表达、酶活性和脂肪代谢,提高肌纤维比例、降低脂肪沉积,改善肉质和胴体品质。This experiment aimed to investigate the effects of rosmarinic acid sweet glycosides(MGE)on muscle energy metabolism enzyme activities,myofibre type composition,and fat metabolism-related gene expression in lake sheep.Forty-five healthy 6-month-old lake sheep were selected and divided into CON group(basal diet),MGE900 group(supplemented with 900 mg/kg MGE),and MGE1200 group(supplemented with 1200 mg/kg MGE)for 56 days.At the end of the experiment,slaughter sampling was conducted to collect subcutaneous fat,longest dorsal muscle,and liver from lake sheep.Succinate dehydrogenase(SDH),lactate dehydrogenase(LDH)and malate dehydrogenase(MDH)activities were measured in the longest dorsal muscle,and myofibre type composition(MYHC I,MYHC IIa,MYHC IIb,and MYHC IIx),myofibre type conversion-related gene PGC-1α,and the expression levels of fat-synthesizable PPARγ,ACC,FAS genes and fat-catabolising ATGL,HSL,and LPL genes in the subcutaneous fat,dorsal longest muscle and liver.The results showed that the expression of MYHC I,MYHC IIa,MYHC IIx and PGC-1αgenes was increased in the MGE group,and SDH activity was increased,while the expression of MYHC IIb genes and LDH activity were decreased.In the MGE group,the expression of PPARγ,ACC,and FAS genes was significantly increased in the longest dorsal muscle,while the expression of PPARγand ACC genes was decreased in the subcutaneous fat and liver.ATGL,HSL,and LPL genes were decreased in dorsal longest muscle expression in MGE group,but increased in subcutaneous fat and liver.The results of the study suggested that rosmarinus officinalis sweet glycosides could regulate muscle gene expression,enzyme activity,and fat metabolism in lake sheep,increase the proportion of muscle fiber,reduce fat deposition,improve meat and carcass quality.
