摘  要:为减少玉米在生产及储存过程中的霉变,本试验筛选出一株对于黄曲霉拮抗作用较强的菌株,通过形态学结合分子生物学确定菌株种属,初步测定菌株的挥发性气体和发酵液对黄曲霉的抑制效果,并采用响应面法优化菌株发酵条件。结果表明:拮抗菌株LK3为暹罗芽孢杆菌(Bacillus siamensis),其挥发性气体和发酵液均能抑制黄曲霉菌丝的生长,且二者对于黄曲霉菌丝生长的抑制作用分别表现为极显著和显著。响应面法优化后菌株LK3的最佳发酵条件为:pH 8、接种量9%、发酵温度35℃、发酵转速205 r/min,优化后OD600较之前提高了37.45%,模型预测结果可靠且优化效果较好。拮抗菌株LK3在玉米生物防霉中具有较好的应用潜力和较高的研究价值。In order to reduce the mildew of corn during production and storage,the experiment screened a strain with an excellent antagonistic effect on Aspergillus flavus.Morphology combined with molecular biology was used for strain identification,and the inhibitory effects of volatile gas and fermentation broth on A flavus were preliminarily measured.Response surface method was used to optimize the fermentation conditions.The results showed that the antagonistic bacterium LK3 was Bacillus siamensis,and its volatile gas and fermentation broth could inhibit the growth of A flavus mycelium,and their inhibitory effects reached significant and extremely significant level,respectively.The optimal fermentation conditions for strain LK3 were pH 8,inoculation amount of 9%,fermentation temperature of 35℃,and fermentation speed of 205 r/min.The optimized OD600 had increased by 37.45%compared with before,and the model prediction results were reliable,the optimization effect was outstanding.The antagonistic bacterium LK3 had good potential for application and high research value in corn antimycotic.
