大蒜素通过调控miR-125b-5p抑制脑出血大鼠炎性因子的机制研究

摘  要:目的探讨大蒜素对脑出血大鼠炎性因子的影响及分子机制。方法实验以胶原酶法制备脑出血大鼠模型,随机分为模型组、大蒜素低、中、高剂量组(15、30、60 mg/kg)、anti-miR-125b-5p组、anti-miR-con组、anti-miR-125b-5p+大蒜素组、anti-miR-con+大蒜素组,另设假手术组。腹腔注射给药24 h后,进行大鼠神经功能评分;实时荧光定量PCR法(qRT-PCR)检测大鼠脑组织中肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素1β(IL-1β)mRNA表达以及miR-125b-5p表达;免疫印迹实验(Western blot)检测大鼠脑组织中TNF-α、IL-6、IL-1β蛋白表达。结果与假手术组比较,模型组神经功能评分、脑组织中TNF-α、IL-6、IL-1βmRNA和蛋白表达明显升高,miR-125b-5p表达明显降低(P<0.05);与模型组比较,大蒜素低、中、高剂量组神经功能评分、脑组织中TNF-α、IL-6、IL-1βmRNA和蛋白表达明显降低,miR-125b-5p表达明显升高(P<0.05)。与anti-miR-con组比较,anti-miR-125b-5p组脑组织中miR-125b-5p表达明显降低,TNF-α、IL-6、IL-1βmRNA和蛋白表达明显升高(P<0.05)。与anti-miR-con+大蒜素组比较,anti-miR-125b-5p+大蒜素组神经功能评分、脑组织中TNF-α、IL-6、IL-1βmRNA和蛋白表达明显升高(P<0.05)。结论大蒜素可能通过上调miR-125b-5p表达抑制脑出血大鼠炎性因子分泌,改善脑出血后神经功能损伤。Objective To explore the effect of allicin on regulating inflammatory factors in rats with intracerebral hemorrhage and its molecular mechanism.Methods The rat model of intracerebral hemorrhage(ICH)was prepared by collagenase method.ICH rats were randomly divided into model group,low-dose,medium-dose and high-dose allicin groups(15,30,60mg/kg),anti-miR-125b-5p group,anti-miR-con group,anti-miR-125b-5p+allicin group,anti-miR-con+allicin group,and sham operation group.After 24 hours of intraperitoneal injection,the neurological function of rats was evaluated.Quantitative real-time polymerase chain reaction(QRT-PCR)was used to detect the mRNA expressions of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β)and miR-125b-5p in rat brain tissue.Western blot was used to detect the protein expressions of TNF-α,IL-6,IL-1βin rat brain tissue.Results Compared with the sham operation group,ICH rats in the model group had significantly higher neurological function score and mRNA and protein levels of TNF-α,IL-6,and IL-1β,but lower mRNA level of miR-125b-5p(P<0.05).Compared with the model group,ICH rats in the low-dose,medium-dose and high-dose allicin groups had significantly lower neurological function score and mRNA and protein levels of TNF-α,IL-6,and IL-1β,but higher mRNA level of miR-125b-5p(P<0.05).Compared with the anti-miR-con group,ICH rats in the anti-miR-125b-5p group had significantly lower mRNA level of miR-125b-5p,but higher mRNA and protein levels of TNF-α,IL-6,and IL-1βin brain tissue(P<0.05).Compared with the anti-miR-con+allicin group,ICH rats in the anti-miR-125b-5p+allicin group had significantly higher neurological function score and mRNA and protein levels of TNF-α,IL-6,and IL-1βmRNA(P<0.05).Conclusion Allicin may inhibit the release of inflammatory factors in ICH rats by up-regulating miR-125b-5p,and thus improves neurological deficits after ICH.
