龙葵多酚提取工艺优化及其抗氧化活性分析

摘  要:以龙葵多酚提取率为指标,经单因素实验和响应面分析法优化其提取工艺,得到最佳工艺条件:乙醇体积分数50%、液料比45∶1(mL/g)、微波温度71℃、微波时间3 min,龙葵多酚提取率2.05%。并对龙葵多酚进行DPPH自由基清除能力实验和羟自由基清除能力实验,以此评价其抗氧化活性。实验结果表明,当龙葵多酚提取液质量浓度为0.036 9 mg/mL时DPPH自由基清除率可达85.01%,当质量浓度为0.589 4 mg/mL时羟自由基清除率56.37%,说明龙葵多酚具有一定的抗氧化活性,可为龙葵资源的开发利用提供参考价值。Taking the extraction rate of solanum nigrum polyphenols as the index,the extraction process was optimized through single factor experiments and response surface analysis.The optimal process conditions were obtained:ethanol volume fraction of 50%,solid-liquid ratio of 45:1(mL/g),microwave temperature of 71℃,microwave time of 3 min,and solanum nigrum polyphenol extraction rate of 2.05%.And DPPH radical scavenging ability and hydroxyl radical scavenging ability experiments were conducted on solanum nigrum polyphenols to evaluate their antioxidant activity.The experimental results showed that the DPPH radical scavenging rate could reach 85.01%when the mass concentration of Solanum nigrum polyphenol extract was 0.0369 mg/mL,and the hydroxyl radical scavenging rate was 56.37%when the mass concentration was 0.5894 mg/mL.This indicates that solanum nigrum polyphenols have certain antioxidant activity and can provide reference value for the development and utilization of solanum nigrum resources.
