摘 要:为探究酶辅助超声提取法中,不同因素对枇杷叶黄酮、三萜酸、多糖提取含量的影响,并研究提取物的生物活性。在单因素试验的基础上,进行响应面优化;采用DPPH和ABTS+·自由基清除试验,分析提取物的抗氧化能力;通过纸片扩散法,探讨其抑菌活性;以过氧化值(POV)、硫代巴比妥酸值(TBARS)、pH值为参考指标,研究提取物对猪肉的保鲜效果。结果表明,最佳提取工艺条件为料液比1∶45(g/mL)、乙醇体积分数65%、超声时间40 min、溶液pH值5.0、复合酶质量添加量1.6%。在此条件下,验证组黄酮、三萜酸、多糖的提取含量分别提高73.66,3.82,4.74 mg/g;在一定的质量浓度范围内,提取物对DPPH和ABTS+·自由基的最高清除率分别达到77.64%和62.41%;提取物对大肠杆菌及巨大芽孢杆菌表现出较好的抑菌效果;经过枇杷叶提取原液浸泡后的猪肉,在保藏过程中,其POV值、TBARS值及pH值明显低于空白对照组。研究为植物性原料活性物质的提取及利用提供参考。In order to study the effects of different factors on the extraction of flavonoids,triterpene acids and polysaccharides in Eriobotrya Folium by ultrasonic enzymatic hydrolysis,and to study the biological activities of the extracts,response surface optimization was carried out on the basis of single factor test.DPPH and ABTS+·free radical scavenging tests were used to analyze the antioxidant capacity of the extracts.Peroxide value(POV),thiobarbituric acid value(TBARS)and pH value were used as reference indexes to study the fresh-keeping effect of the extract on pork.The results show that the optimal extraction conditions are as follows:The ratio of solid to liquid is 1∶45(g/mL),the volume fraction of ethanol is 65%,the ultrasonic time is 40 min,the pH value of solution is 5.0,the addition amount of complex enzyme is 1.6%.Under these conditions,the extraction contents of flavonoids,triterpenoid acid and polysaccharide in the verification group are increased by 73.66 mg/g,3.82 mg/g and 4.74 mg/g,respectively.In a certain mass concentration range,the highest scavenging rates of DPPH and ABTS+·free radicals are 77.64%and 62.41%.The results of bacteriostatic experiment show that the extract has good bacteriostatic effect on Escherichia coli and Bacillus gigantium.The POV,TBARS and pH values of the pork soaked in loquat leaf extract are significantly lower than those of the control group during storage.In summary,this study provides a certain reference for the extraction and utilization of active substance from plant raw materials.
