胃癌中semaphorin 5A和PD-L1的表达与临床病理的相关性

摘　　要：目的探讨胃癌中神经轴突导向分子5A(semaphorin 5A,SEMA5A)和程序性死亡配体-1(programmed death ligand-1,PD-L1)的表达及临床意义。方法收集41例胃癌组织与配对的癌旁组织临床资料,采用免疫组化法和RT-qPCR法检测SEMA5A、PD-L1的表达,并分析SEMA5A、PD-L1的相关性。应用慢病毒构建SEMA5A稳定低表达细胞系,运用RT-qPCR、Western blot法分析SEMA5A和PD-L1在胃癌细胞中表达的相关性。结果免疫组化染色显示胃癌、癌旁组织中SEMA5A蛋白的高表达率分别为65.9%(27/41)、39.0%(16/41),PD-L1蛋白阳性率分别为58.5%(24/41)、14.6%(6/41)。RT-qPCR检测胃癌、癌旁组织中SEMA5A mRNA相对表达量为1.30±0.50、0.81±0.48,PD-L1 mRNA相对表达量为0.70±0.42、0.12±0.09。SEMA5A表达与胃癌组织学分型、淋巴结转移相关(P<0.05);PD-L1表达与肿瘤大小、T分期、胃癌病理分期相关(P<0.05)。Pearson相关分析示胃癌组织中SEMA5A和PD-L1 mRNA的表达呈正相关,Spearman相关性分析示癌旁组织中两者表达无相关性。敲低人胃腺癌细胞株SEMA5A mRNA后,PD-L1表达下调。结论SEMA5A和PD-L1在胃癌组织中高表达,两者有相关性,可作为胃癌预后评估和联合治疗的潜在分子标志物。Purpose To investigate the expression of semaphorin 5A(SEMA5A)and programmed death ligand-1(PD-L1)and their clinicopathological significance in gastric cancer.Methods Clinical data of 41 cases of gastric cancer tissues and paired adjacent tissues were collected.Immunohistochemical staining and RT-qPCR were used to detect the expression levels of SEMA5A and PD-L1,and analysed the correlation between SEMA5A and PD-L1 and clinicopathological features.In addition,we used lentivirus to construct SEMA5A stable low-expression cell lines.RT-qPCR and Western blot were used to analyse the correlation between the expression of SEMA5A and PD-L1 in gastric cancer tissues.Results The high expression rate of SEMA5A was 65.9%(27/41)in gastric cancer tissues and 39.0%(16/41)in paracancerous tissues,respectively.The positive rates of PD-L1 were 58.5%(24/41)and 14.6%(6/41),respectively.RT-qPCR showed that the relative expression levels of SEMA5A mRNA in gastric cancer and paracancerous tissues were 1.30±0.50 and 0.81±0.48,respectively,while the relative expression levels of PD-L1 mRNA were 0.70±0.42 and 0.12±0.09,respectively.SEMA5A expression was correlated with histological typing of gastric cancer and lymph node metastasis(P<0.05).PD-L1 expression was correlated with tumour size,T stage,and pathological stage of gastric cancer(P<0.05).Pearson correlation analysis showed a positive correlation between the expression of SEMA5A and PD-L1 mRNA in gastric cancer tissues,and spearman correlation analysis showed that there was no correlation between the expression of the two in paracancerous tissues.Knockdown of SEMA5A gene in human gastric adenocarcinoma cell lines resulted in down-regulation of PD-L1 expression.Conclusion Both SEMA5A and PD-L1 are highly expressed in gastric cancer tissues,and there is a correlation between the expressions of SEMA5A and PD-L1.They can serve as potential molecular markers for prognostic evaluation and combination therapy of gastric cancer.
