摘  要:目的以奶西姆提、砀山梨和鸭梨三个不同品种梨为主要原料,通过优化碱液浓度、碱解温度、碱解时间等因素,优化膳食纤维提取工艺,对提取的可溶性膳食纤维进行抗氧化活性测定。方法通过正交试验分析碱液浓度、碱解温度、碱解时间对膳食纤维提取率的影响,从而确定最佳提取工艺。同时利用DPPH自由基、ABTS+自由基、羟基自由基的清除能力和三价铁离子的还原能力来测定可溶性膳食纤维的抗氧化活性。结果粗纤维含量大小依次为奶西姆提、砀山梨和鸭梨、;膳食纤维含量大小依次为奶西姆提、砀山梨、鸭梨;对DPPH自由基、ABTS+自由基、羟基自由基的清除率和对三价铁离子的还原能力大小依次为奶西姆提、砀山梨、鸭梨。结论奶西姆提膳食纤维的最佳提取条件为碱解温度70℃、碱解时间80 min、碱液浓度7%,在此条件下其提取率为8.47%;随着浓度的升高,清除率和还原能力升高,抗氧化活性提高,为其膳食纤维在体外细胞实验的研究提供了初步基础。Objective Taking three different varieties of pears,namely,Korla fragrant pears,Dangshan pears and duck pears,as the main raw materials,the dietary fiber extraction process was optimized through orthogonal experiments,and the antioxidant activity of the extracted soluble dietary fiber was determined.Methods The effects of three factors on the extraction rate of dietary fiber were analyzed by orthogonal test to determine the optimal extraction process.The antioxidant activity was also determined by the in vitro free radical scavenging ability and the reducing ability of trivalent iron ions.Results The size of crude fiber content was in the order of Dangshan pear,duck pear,and Korla fragrant pear;the size of dietary fiber content was in the order of Korla fragrant pear,Dangshan pear,and duck pear;the size of in vitro antioxidant activity was in the order of Korla fragrant pear,Dangshan pear,and duck pear.Conclusions The optimal extraction conditions of dietary fiber of Korla balsam pear were alkalinity temperature 70℃,alkalinity time 80 min,alkaline concentration 7%,and the extraction rate was 8.47%under this condition;the scavenging rate of DPPH radicals,ABTS+radicals,hydroxyl radicals,and the reducing ability of trivalent iron ions had a quantitative and effective relationship,which provided a theoretical basis for further research on its dietary fiber.
