摘 要:以葡萄柚皮为原料,采用水热法制备碳量子点(G-CQDs),并以此构建一种用于金银花中Pb^(2+)检测的荧光生物传感器。利用圆二色光谱、透射电子显微镜、紫外-可见吸收光谱、荧光光谱等对G-CODs的形貌和光学性能进行表征,优化该荧光生物传感器的实验条件,并对其检测性能进行分析。结果表明:G-CQDs呈球形且分布均匀,平均粒径为2.41 nm,最佳激发波长为350 nm;荧光生物传感器的适宜实验条件为Hemin浓度60μmol/L、反应时间30 min、反应温度25℃、K^(+)浓度10 mmol/L和pH值为6.5;与其他金属离子(Ag^(+)、Zn^(2+)、Cd^(2+)、Cu^(2+)和Hg^(2+))相比,荧光生物传感器对Pb^(2+)的选择性更高,且其Δ荧光强度随着Pb^(2+)质量浓度的增加而增强,在0.1~5.0μg/mL线性范围内,二者线性关系良好,R 2为0.9980,检测限为0.0639μg/mL;与传统方法相比,该荧光生物传感器的准确性良好,且具有绿色环保、经济有效、操作简单等优势。Carbon quantum dots(G-CQDs)were prepared by hydrothermal method using grapefruit peel and fluorescence biosensor for Pb^(2+)detection was constructed in honeysuckle.The morphology,structure and optical properties of G-CODs were characterized by circular dichroism spectroscopy,transmission electron microscopy,ultraviolet-visible absorption spectroscopy and fluorescence spectroscopy.The experimental conditions of the fluorescent biosensor were optimized and its detection performance was analyzed.The results showed that the G-CQDs were spherical and uniformly distributed.The average particle size was 2.41 nm,and the optimal excitation wavelength was 350 nm.The suitable experimental conditions for the fluorescence biosensor were as follows:Hemin concentration of 60μmol/L,reaction time of 30 min,reaction temperature of 25℃,K^(+)concentration of 10 mmol/L and pH value of 6.5.Compared with other metal ions(Ag^(+),Zn^(2+),Cd^(2+),Cu^(2+)and Hg^(2+)),the fluorescence biosensor had higher selectivity for Pb^(2+),and its fluorescence intensity increased with the increase of Pb^(2+)mass concentration.In the linear range of 0.1~5.0μg/mL,the linear relationship between the two was good,R 2 was 0.9980,and the detection limit was 0.0639μg/mL.Compared with the conventional method(ICP-MS method),the fluorescence biosensor had good accuracy,and had the advantages of green environmental protection,economical and effective,simple operation and so on.
