摘 要:试验旨在研究不同蛋白酶对魔芋飞粉的酶解效果。采用液态法酶解魔芋飞粉,以酶解液中多肽含量为指标,采用单因素试验、正交试验优化不同蛋白酶(木瓜蛋白酶、中性蛋白酶、酸性蛋白酶、碱性蛋白酶、菠萝蛋白酶)的酶解工艺,对比最优酶解条件下不同蛋白酶的酶解液中多肽含量。结果显示,不同蛋白酶的最适条件、酶解效果均存在差异,通过优化酶解工艺能够显著提高酶解效果。酸性蛋白酶的对魔芋飞粉的酶解效果优于其他4种蛋白酶,最优酶解条件为酶解温度65℃、pH值1.5、酶解时间3.0 h、加酶量3600 U/g,其酶解液中多肽含量为7.51g/L。研究表明,酸性蛋白酶酶解有助于实现魔芋飞粉在饲料产业中的高值化利用。The study aimed to investigate the enzymolysis effects of different proteases on konjac fly powder.The liquid method was used to enzymolysis konjac fly powder.Single factor and orthogonal tests were used to optimize the enzymolysis process of different proteases(papain,neutral,acid,alkaline,and bromelain)based on the polypeptide content in the enzymolysis solution.The polypeptide contents of different proteases in the enzymolysis solution were compared under the optimal enzymolysis conditions.The results showed that the optimum conditions and enzymolysis effects of different proteases were different,and the enzymolysis impact could be improved significantly by optimizing the enzymolysis process.The enzymolysis effect of acid protease on konjac fly powder was better than that of the other four proteases.The optimal enzymolysis conditions of acid protease were enzymolysis temperature of 65℃,pH value of 1.5,enzymolysis time of 3.0 h,enzymolysis amount of 3600 U/g,and the polypeptide content of the enzymolysis solution was 7.51 g/L.The study indicates that enzymatic hydrolysis using acid protease can facilitate the high-value utilization of konjac powder in the feed industry.
