摘  要:目的探究镰形棘豆的体外抗氧化和抑菌活性部位,并采用超高效液相色谱-串联质谱(UPLC-MS/MS)法对高活性部位中的化学成分进行鉴定。方法采用定性鉴别反应初步分析镰形棘豆的水提取物(WTE)、乙酸乙酯提取物(EAE)和无水乙醇提取物(ETE)的组成;测定DPPH、ABTS自由基清除能力以评估各提取物的抗氧化能力;利用倍比稀释法测定提取物对金黄色葡萄球菌、铜绿假单胞菌的抑菌效果,筛选出最佳抗氧化和抑菌活性部位。应用UPLC-MS/MS技术,鉴定高活性部位的化学成分。结果WTE、EAE和ETE对DPPH和ABTS自由基的清除能力均呈剂量依赖性增加,其中ETE具有最强的抗氧化活性,其半抑制-1浓度(IC50)分别为0.41 mg·mL和0.08 mg·mL^(-1)。EAE和ETE均对金黄色葡萄球菌具有较好的抑菌效果,其最小抑菌浓度(MIC)均为6.25mg·mL^(-1),最小杀菌浓度(MBC)均为25.00 mg·mL^(-1),对铜绿假单胞菌无抑菌活性;而WTE对金黄色葡萄球菌和铜绿假单胞菌均无抑菌效果。从ETE的质谱数据分析出92个化学成分,包含黄酮类58个,生物碱类16个,三萜类7个和其他类化合物11个。其中11个黄酮类、5个生物碱类、2个三萜类和10个其他类化合物在镰形棘豆中为首次报道。结论富含黄酮类物质的ETE表现出最佳的抗氧化和抑菌活性;建立的UPLC-MS/MS法可对镰形棘豆中化学成分进行准确、快速、全面的分析。Objective To determine the antioxidant and antibacterial activity of Oxytropis falcata Bunge(OFB)and to identify the chemical constituents of the most active fraction by ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Methods The compositions of the water extract(WTE),ethyl acetate extract(EAE),and anhydrous ethanol extract(ETE)of OFB were analyzed by qualitative identification reactions.Antioxidant capacity was assessed by the scavenging ability of DPPH and ABTS radicals.The antibacterial effect of the extract against Staphylococcus aureus(S.aureus)and Pseudomonas aeruginosa(P.aeruginosa)was determined with broth dilution method.UPLC-MS/MS was used to identify the chemical components of the highly-active parts.Results The scavenging abilities of WTE,EAE,and ETE against DPPH and ABTS radicals increased in a dose-dependent manner,and ETE demonstrated the-1 highest antioxidant activity with the inhibitory concentration 50(IC50)at 0.41 mg·mL and 0.08 mg·mL^(-1),respectively.Antibacterial experiment indicated that both EAE and ETE had certain-1 bacteriostatic effects on S.aureus,the minimum inhibitory concentration(MIC)was 6.25 mg·mL^(-1) for all,and the minimum bactericidal concentration(MBC)was 25.00 mg·mL for all.However,neither showed antibacterial activity against P.Aeruginosa,and WTE lacked antibacterial effect on S.aureus and P.Aeruginosa.From the ETE mass spectrometry data,92 chemical components were identified,including 58 flavonoids,16 alkaloids,7 triterpenoids,and 11 other compounds.Notably,11 flavonoids,5 alkaloids,2 triterpenoids,and 10 other compounds were reported for the first time in the Oxytropis falcata Bunge.Conclusion ETE rich in flavanoids shows strongest antioxidant and antibacterial activities,and the UPLC-MS/MS method is accurate and rapid for the comprehensive analysis of the chemical components in Oxytropis falcata Bunge.
