摘　　要：目的 结合科研与教学实践,设计一个利用杂合优化及毕赤酵母外源表达技术高效制备新型海洋重组抗菌肽的综合实验。方法 采用生物信息学方法对数据库中海洋来源抗菌肽进行理化性质分析,获得科研价值高、应用潜力大的候选序列;经杂合优化后全基因合成,再转入毕赤酵母中重组表达,通过滤纸片琼脂扩散法进行活性评价。结果 筛选到1株毕赤酵母工程菌,能高效表达抑菌活性较好的牡蛎杂合抗菌肽Def-Prp。结论 该设计结合了多学科实验技能,锻炼了学生科研创新思维和分析解决问题的能力,也为重组抗菌肽的开发利用提供了一条可资借鉴的技术途径。Objective To design a comprehensive experiment using hybrid optimization and Pichia pastoris exogenous expression technology through combination of scientific research and teaching practice to efficiently prepare a novel marine recombinant antimicrobial peptide.Methods Bioinformatics methods were used to analyze the physicochemical properties of marine-derived antimicrobial peptides in the database,and candidate sequences with high scientific research value and great application potential were obtained.After hybrid optimization,the whole gene was synthesized and then transferred into Pichia pastoris for recombinant expression.The activity was evaluated by filter paper agar diffusion method.Results The results showed that one engineered strain of Pichia pastoris was screened,which could efficiently express the oyster hybrid antimicrobial peptide Def-Prp with good antibacterial activity.Conclusion The design combines multidisciplinary experimental skills,exercises students' innovative thinking in scientific research and the ability to analyze and solve problems,and also provides a technical way for the development and utilization of recombinant antimicrobial peptides.
