摘  要:目的采用孟德尔随机化(MR)方法评估19种营养素与乳腺癌之间的潜在因果关系。方法获取19种营养素的全基因组关联研究(GWAS)数据和芬兰数据库R11版本中乳腺癌的全基因组关联分析数据。采用逆方差加权(IVW)、MR-Egger回归法、加权中位数、加权模型、简单模型5种方法研究19种营养素和乳腺癌之间的因果关系。使用Cochran’s Q检验进行异质性分析,并使用MR-Egger回归法进行水平多效性评估。结果5种MR分析结果均显示,铜、钙、β-胡萝卜素、叶酸、铁、镁、钾、硒、维生素A、维生素B12、维生素B6、维生素C、维生素D、维生素E、锌、碳水化合物、脂肪、蛋白质、全糖19种营养素与乳腺癌无因果关系(P﹥0.05),敏感性分析均未发现存在显著异质性和水平多效性(P﹥0.05)。结论从基因驱动的角度来看,19种营养素与乳腺癌之间不存在因果关系,需要更多的研究进一步明确它们之间的关系。Objective To evaluate the potential causal relationship between 19 nutrients and breast cancer by Mendelian randomization(MR).Method The genome-wide association study(GWAS)data for 19 nutrients and the genomewide association analysis data for breast cancer in the R11 version of the Finnish database were obtained.The inverse variance weighted(IVW),MR-Egger regression,weighted median,weighted model and simple model were used to study the causal relationship between 19 nutrients and breast cancer.Cochran’s Q test was used for heterogeneity analysis,and MR-Egger regression was used for horizontal pleiotropy assessment.Result All five MR analysis results showed that 19 nutrients,including copper,calcium,beta-carotene,folate,iron,magnesium,potassium,selenium,vitamin A,vitamin B12,vitamin B6,vitamin C,vitamin D,vitamin E,zinc,carbohydrate,fat,protein,and whole sugar,had no causal relationship with breast cancer(P>0.05),no significant heterogeneity or horizontal pleiotropy was found in sensitivity analysis(P>0.05).Conclusion From a gene drive perspective,there is no causal relationship between the 19 nutrients and breast cancer,and more researches are needed to further clarify the relationship between them.
