摘 要:采用超声辅助乙醇-硫酸铵双水相体系从菜籽饼中提取菜籽多酚。在单因素实验基础上,采用响应面法优化提取工艺条件,以抗坏血酸为阳性对照,研究菜籽多酚对DPPH·清除率、·OH清除率、ABTS^(+)·清除率和总抗氧化能力,综合评价菜籽多酚体外抗氧化活性。结果表明:菜籽多酚最佳提取工艺参数为乙醇体积分数33%,料液比1:50,超声时间40 min,该条件下总多酚提取率为6.9600 mg·g^(-1),与回归模型预测值(7.0240 mg·g^(-1))接近,该工艺稳定可行。所得菜籽多酚对DPPH·、·OH、ABTS^(+)·的清除能力和总抗氧化能力均随浓度增高而增强,IC_(50)分别为1.75μg·mL^(-1)、194.00μg·mL^(-1)、3.05μg·mL^(-1),表现出良好的抗氧化活性。Rapeseed polyphenols were extracted from rapeseed cake by ultrasonic-assisted ethanol-ammonium sulfate aqueous system.On the basis of single factor experiment,the Box-Behnken response surface method was used to optimize the extraction process conditions,and ascorbic acid was used as positive control to study the removal rate of rapeseed polyphenols on DPPH·clearance,·OH clearance,ABTS^(+)·clearance and total antioxidant capacity,and comprehensively evaluate the antioxidant activity of rapeseed polyphenols in vitro.The results showed that the optimal extraction parameters of rapeseed polyphenols were ethanol volume fraction 33%,solid-liquid ratio 1:50,and ultrasonic time 40 min.Under this conditions,the extraction rate of total polyphenols was 6.9600mg·g^(-1),which was close to the predicted value of the regression model(7.0240 mg·g^(-1)).The process was stable and feasible.The scavenging ability of rapeseed polyphenols to DPPH·,·OH and ABTS^(+)·,and the total antioxidant capacity were enhanced with increasing concentration,with IC_(50)of 1.75μg·mL^(-1),194.00μg·mL^(-1)and 3.05μg·mL^(-1),showing good antioxidant activity.
