摘 要:目的分析KAI1/CD82、p53蛋白在胃癌发生、发展机制中的调控作用。方法体外培养胃癌SGC7901细8胞,按照实验要求分为阴性对照组[加入等体积的0.9%NaCl]、A组[rAd-p53低剂量组(浓度为5×10 vp/mL)]、B组108[rAd-p53高剂量组(浓度为5×10 vp/mL)]、C组[DDP组(浓度为2.5 mg/L)]、D组[低剂量联合组(浓度为5×10 vp/mL+2.510 mg/L)]、E组[高剂量联合组(5×10 vp/mL+2.5 mg/L)]。rAd-p53、DDP低剂量组、DDP高剂量组及DDP低高剂量联合组作用于细胞株48 h后,采用免疫组化SP法和Western-blot法检测并比较不同组别间的KAI1/CD82、p53蛋白表达水平,采用Pearson相关分析法分析SGC-7901细胞中KAI1/CD82与p53蛋白的表达的相关性。选取2020年6月至2021年6月期间于海口市人民医院胃肠外科实施手术的43例胃癌切除术后患者的标本,将标本分为胃癌组、癌旁组(距离癌灶2 cm)和正常组织组(距离癌灶5 cm以上)。采用Western blot法检测三组样本中的KAI1/CD82和p53蛋白表达水平,并比较不同临床病理特征患者胃癌组织中KAI1/CD82、p53蛋白的表达水平,分析其与胃癌的临床病理特征的关系。结果免疫组化法检测结果显示,与阴性对照组比较,A组和B组细胞中KAI1/CD82、p53蛋白的阳性表达率均显著升高,差异均有统计学意义(P<0.05);D组与B组比较,KAI1/CD82蛋白表达上调,p53蛋白表达进一步升高,差异均有统计学意义(P<0.05);Western blot法检测结果显示,rAd-p53单独及联合DDP作用48 h后,SGC-7901细胞中KAI1/CD82、p53蛋白的表达量均高于阴性对照组,且低高剂量联合组(D、E组)中KAI1/CD82、p53蛋白表达也明显高于低高剂量单药组(A、B组),差异均有统计学意义(P<0.05);Pearson相关分析结果显示,SGC-7901细胞中KAI1/CD82与p53蛋白的表达水平呈正相关(r=0.962,P<0.05)。Western-blot法分析结果显示,胃癌组织中KAI1/CD82和p53蛋白的表达水平明显低于其在癌旁组织和正常组织中的表达水平,癌旁组织�Objective To analyze the regulatory roles of KAI1/CD82 and p53 proteins in the mechanisms of gastric cancer occurrence and development.Methods Gastric cancer SGC7901 cells were cultured in vitro and divided into the following groups according to experimental requirements:negative control group(equal volume of 0.9%NaCl),group A(low-dose rAd-p53 group,5×108 vp/mL),group B(high-dose rAd-p53 group,5×1010 vp/mL),group C(DDP group,2.5 mg/L),group D(low-dose combination group,5×108 vp/mL rAd-p53+2.5 mg/L DDP),and group E(high-dose combination group,5×1010 vp/mL rAd-p53+2.5 mg/L DDP).After 48 hours of treatment with rAd-p53,low-/high-dose DDP,or their combinations,the expression levels of KAI1/CD82 and p53 proteins in each group were detect-ed and compared using immunohistochemical SP and Western blot methods.Pearson correlation analysis was performed to assess the relationship between KAI1/CD82 and p53 protein expression in SGC-7901 cells.Surgical specimens from 43 gastric cancer patients who underwent resection at the Department of Gastrointestinal Surgery,Haikou People's Hospi-tal(from June 2020 to June 2021)were collected and categorized into gastric cancer tissue,adjacent tissue(2 cm from the tumor),and normal tissue(≥5 cm from the tumor).Western blot was used to measure KAI1/CD82 and p53 protein expression levels in these tissues.Differences in protein expression across clinicopathological features were analyzed,along with their correlations.Results Immunohistochemistry showed significantly higher positive expression rates of KAI1/CD82 and p53 proteins in groups A and B compared to the negative control(P<0.05).Group D exhibited upregulat-ed KAI1/CD82 and further elevated p53 expression versus group B(P<0.05).Western blot confirmed that rAd-p53 alone or combined with DDP increased KAI1/CD82 and p53 expression in SGC-7901 cells versus the control,with combination groups(D,E)showing higher levels than monotherapy groups(A,B),P<0.05.Pearson analysis revealed a positive correla-tion between KAI1/CD82 and p53 expre
