摘 要:生物钟是机体为适应昼夜更替而形成的一种内在节律性,生物钟紊乱会严重影响机体健康。随着“肠-脑轴”理论发展,益生菌被证明可能通过分泌神经递质调节生物钟紊乱。褪黑素(Melatonin,MT)主要由神经内分泌器官松果体合成,具有调节生物钟的功能。文章对前期筛选得到的1株高产MT短乳杆菌SLB2(Levilactobacillus brevis SLB2)进行益生特性评价,探究其对小鼠生物钟紊乱的改善作用。结果表明,短乳杆菌SLB2具有良好益生特性,能够回调生物钟紊乱小鼠摄食量及体质量,增加小鼠在旷场试验中的中央区域停留时间,提升小鼠在新物体识别试验中的学习能力,增加小鼠在明暗箱试验中的明箱停留时间及明箱移动距离,焦虑现象得到明显改善,预防由于生物钟紊乱导致的结肠炎症发生;其作用机制与显著增加血清和结肠中的5-羟色胺(5-HT)、MT含量(P<0.05),防止结肠组织局部淋巴细胞浸润,增加肠道菌群厚壁菌门丰富度有关,为进一步开发改善生物钟紊乱的益生菌制剂提供理论依据。The biological clock is an intrinsic rhythmicity formed by the organism to adapt to the change of day and night.Disorders of the biological clock can seriously affect the health of the organism.With the development of the‘gut-brain axis’theory,probiotics have been shown to regulate biological clock disorders by secreting neurotransmitters.Melatonin(MT)is mainly synthesised by the pineal gland,a neuroendocrine organ,and has the function of regulating the biological clock.In this study,a high MT-producing strain of Levilactobacillus brevis SLB2 obtained from a pre-screening,was evaluated for its probiotic properties and its ameliorative effect on biological clock disorders in mice was investigated.The results showed that Levilactobacillus brevis SLB2 has good probiotic properties,which can regulate the intake and body weight of mice with biological clock disorders,increase the residence time in the central area of mice in the open field experiment,improve the learning ability of mice in the new object recognition experiment,increase the residence time and the distance of mice in the bright box in the bright and dark boxes experiment,improve the anxiety phenomenon,and prevent the occurrence of colon inflammation due to the disorders of the biological clock.The mechanism of action is related to the significant increase in serum and colonic levels of 5-HT and MT(P<0.05),the prevention of local lymphocyte infiltration in colonic tissues,and the increase in the abundance of intestinal flora.This study provides a theoretical basis for further development of probiotic preparations to improve biological clock disorders.
