摘  要:氧化应激损伤会导致衰老和各种疾病,因此开发天然抗氧化剂具有重要意义。对小叶紫薇茎叶器官不同萃取部位的抗氧化活性及活性成分进行对比研究,以期发现潜在药源。将小叶紫薇茎叶乙醇提取物依次用石油醚(PE)、乙酸乙酯(EAC)和正丁醇(NBA)萃取,采用紫外分光光度法检测不同样品中活性成分黄酮、酚酸含量。对样品进行1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二胺盐(ABTS)抗氧化的反应动力学研究,考察反应时间对样品清除自由基效果的影响,寻找合适的反应终点,以确保抗氧化检测结果的准确性;进一步采用DPPH、ABTS+自由基清除实验对样品的体外抗氧化能力进行评价。结果显示小叶紫薇茎叶不同萃取部位中,黄酮和酚酸含量依次为EAC-E>NBA-E>PE-E。反应动力学研究结果表明,在样品的DPPH、ABTS+自由基清除实验中,30 min为反应达到平衡所需时间。体外抗氧化实验结果表明,乙酸乙酯萃取物(EAC-E)活性最强,与阳性对照叔丁基对苯二酚(TBHQ)接近;石油醚萃取物(PE-E)最弱。小叶紫薇茎叶中活性成分黄酮、酚酸的含量与抗氧化活性呈正相关。该研究表明小叶紫薇茎叶不同萃取部位中EAC-E的活性成分及抗氧化活性最高,可用于天然抗氧化剂的开发,为小叶紫薇废弃资源茎叶的综合利用提供科学的理论依据。Oxidative stress damage can lead to aging and various diseases.Therefore,it is of great significance to develop natural antioxidants.A comparative study was conducted on the antioxidant activity and active component of different extraction parts of the stem and leaf of Lagerstroemia indica,with the expectation of discovering potential sources of medicine.The ethanol extract of the stem and leaf of Lagerstroemia indica was extracted with petroleum ether(PE),ethyl acetate(EAC),and n-butanol(NBA).The contents of flavonoids and phenolic acids in different samples were determined by UV spectrophotometry.The reaction kinetics of DPPH and ABTS+radical scavenging assays was studied,and the effect of reaction time on the scavenging effect of free radicals was investigated.The appropriate reaction end point was found to ensure the accuracy of antioxidant test results.Further more,The DPPH·and ABTS+·scavenging capacity were used to evaluate the antioxidant capacity in vitro of the samples.The results showed that among the different extraction parts of the stems and leaves of Lagerstroemia indica,the contents of flavonoids and phenolic acids were in the order of EAC-E>NBA-E>PE-E.It could be seen from the results of reaction kinetics that 30 min was the time required for the reaction to reach equilibrium in the DPPH·,ABTS+·scavenging experiments.The results of in vitro antioxidant tests showed that the ethyl acetate fraction(EAC-E)had the strongest activity and was close to the positive control TBHQ,while the petroleum ether fraction(PE-E)had the weakest activity.The contents of flavonoids and phenolic acids in the stem and leaf of Lagerstroemia indica were positively correlated with their antioxidant activity.The study showed that the active components and antioxidant activities of EAC-E were the highest in different extracts from the stem and leaf of Lagerstroemia indica,which could be used for the development of natural antioxidants.It will provide a scientific theoretical basis for the comprehensive utilization of the
