摘  要:采用三级组合膜方法(超滤、纳滤和反渗透相结合),以水为溶剂,从刺梨中富集小分子抗氧化活性成分。从5种不同截留分子量的聚乙烯膜中筛选出较合适的超滤膜,以多糖透过率、维生素C(vitamin C,V_(C))透过率和平均膜通量为指标,在单因素试验的基础上结合响应面分析法优化超滤工艺参数;以酪氨酸酶抑制率、DPPH自由基和ABTS+自由基清除率为指标,筛选纳滤膜,并通过单因素试验优化纳滤工艺参数;以反渗透膜浓缩纳滤透过液,制备富含V_(C)的小分子抗氧化提取物,最后采用液相色谱串联-质谱(liquid chromatography-tandem mass spectrometry,LCMS/MS)技术分析提取物的化学组成并采用高效液相色谱(high performance liquid chromatography,HPLC)法测定V_(C)含量,探讨组合膜分离对活性成分的影响。结果表明:优化后的工艺条件为5 kDa超滤膜、500 Da纳滤膜和反渗透膜相结合。超滤参数为刺梨汁稀释20倍、跨膜压力0.4 MPa、过滤体积6 L、冲洗1次;纳滤操作压力为0.8 MPa、冲洗3次。反渗透膜浓缩后,V_(C)得率最高,为88.90%,浓缩液经冷冻干燥得到具有抗氧化活性的小分子天然产物。A three-tiered membrane filtration method,combining ultrafiltration,nanofiltration,and reverse osmosis,was used to enrich small molecular antioxidant components from Rosa roxburghii Tratt with water as the solvent.The appropriate ultrafiltration membrane was selected from polyethylene membranes with five different molecular weight cut-offs.With the polysaccharide permeation rate,vitamin C(V_(C))permeation rate,and membrane flux as indicators,response surface methodology and single factor experiments were employed to optimize the ultrafiltration parameters.The nanofiltration membrane was screened with the tyrosinase inhibition rate,DPPH radical scavenging activity,and ABTS+radical scavenging activity as indicators,and nanofiltration parameters were optimized by single factor experiments.Finally,the nanofiltration permeate was concentrated with the reverse osmosis membrane to prepare the small molecular antioxidant extract rich in V_(C).The chemical composition of the extract was analyzed by liquid chromatography-tandem mass spectrometry(LCMS/MS),and the V_(C) content was determined by high-performance liquid chromatography(HPLC),on the basis of which the impacts of membrane separation on active components were studied.The results showed that the optimized conditions were a combination of a 5 kDa ultrafiltration membrane,a 500 Da nanofiltration membrane,and a reverse osmosis membrane.The ultrafiltration parameters were dilution of R.roxburghii juice by 20 times,transmembrane pressure of 0.4 MPa,filtration volume of 6 L,and flushing once.The nanofiltration parameters were pressure of 0.8 MPa and three flushes.After concentration by the reverse osmosis membrane,the highest yield of V_(C) was 88.90%,and the small molecular natural products with antioxidant activity were obtained after freeze-drying.
