摘 要:目的探讨虾青素对非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)的潜在治疗作用机制。方法利用油酸钠诱导人正常肝细胞(THLE-2)成非酒精性脂肪肝细胞模型。利用虾青素、铁死亡激活剂(Erastin)处理细胞模型,并过表达SLC7A11,检测细胞脂肪,细胞增殖,炎症(IL-6、TNF-α),氧化应激(线粒体膜电位、MDA、GSH-Px)和铁死亡(HMOX1、GPX4)水平。结果与对照组相比,模型组和Erastin处理降低了细胞增殖,提高了氧化应激、炎症以及铁死亡水平(P<0.001);与模型组比较,虾青素处理显著促进了细胞增殖,抑制了氧化应激、炎症以及铁死亡水平升高(P<0.01);虾青素处理和过表达SLC7A11后部分减弱了Erastin的影响(P<0.05)。结论虾青素处理显著改善了非酒精性脂肪肝细胞模型的炎症和铁死亡水平,从而在NAFLD中发挥治疗作用。Objective To explore the potential therapeutic mechanisms of astaxanthin in the treatment of non-alcoholic fatty liver disease(NAFLD).Methods Human normal liver cells(THLE-2)were induced into a NAFLD cell model using sodium oleate.The cell model was treated with astaxanthin,a ferroptosis activator(Erastin),and an overexpression of SLC7A11.Various cellular parameters were assessed,including lipid accumulation,cell proliferation,inflammation(IL-6,TNF-α),oxidative stress(mitochondrial membrane potential,MDA,GSH-Px)and ferroptosis(HMOX1,GPX4).Results Compared with the control group,the model group and Erastin treatment reduced cell proliferation,increased oxidative stress,higher inflammation and elevated ferroptosis markers(P<0.001).In contrast,astaxanthin treatment significantly promoted cell proliferation,reduced oxidative stress and inflammation,and enhanced ferroptosis levels compared to the model group(P<0.01).Furthermore,astaxanthin treatment and SLC7A11 overexpression partially mitigated the effects of Erastin(P<0.05).Conclusion Astaxanthin treatment significantly improved inflammation and ferroptosis levels in NAFLD cell model,thereby exerting therapeutic effects in NAFLD.
