摘  要:目的:本研究旨在利用大规模全基因组关联研究(genome-wide association study,GWAS)结果汇总数据,探讨吸烟行为与肠道微生物之间的遗传相关性和因果关系。方法:选择相互独立且与吸烟行为及肠道微生物组相关的遗传位点作为工具变量。应用逆方差加权法(inverse variance weighted,IVW)及加权中位数法进行孟德尔随机化分析(mendelian randomization,MR),Cochran Q检验评估单核苷酸多态性(single nucleotide polymorphism,SNP)间的异质性;MR多效性残差以及异常值检测SNP异常值;MR-Egger截距测试检验SNP的水平多效性;留一法敏感性分析检验孟德尔随机化研究分析验证研究结果的稳健性。结果:孟德尔随机化研究表明,克里斯滕森菌科(family.Christensenellaceae)与开始吸烟年龄(smoking initiation,SmkInit)之间存在单向的因果关系。结论:肠道微生物应作为影响SmkInit的一个重要因素,为理解吸烟行为及其对精神健康的影响提供了新的视角。Objective:The aim of this study was to use genome-wide association study(GWAS)to aggregate statistical data to examine genetic correlations and causation between smoking and microbiota.Method:The independent genetic loci associated with smoking and the gastrointestinal microbiome were selected as instrumental variables.Inverse variance weighted(IVW)and weighted median methods were applied to Mendelian randomization(MR).Cochran Q test was used to assess heterogeneity among single nucleotide polymorphisms(SNP).MR pleiotropy residual and outlier were used to detect of SNP outlier.MR Egger intercept test were used to test the horizontal polymorphism of SNP.A leave-one-out sensitivity analysis was performed to validate the robustness of the MR study results.Results:Mendelian randomization analysis revealed a unidirectional causal relationship between family.Christensenellaceae and smoking initiation(SmkInit).Conclusion:The gut microbiome should be considered as an important factor influencing SmkInit,providing new perspectives for understanding the impact of smoking behavior on mental health.
