摘  要:为解析火龙果Selenicereus undatus叶绿体基因组结构和序列特征,并探讨其系统进化关系,运用高通量测序技术开展火龙果叶绿体基因组测序,使用生物学信息技术对其组装和注释,分析其密码子偏好性、简单重复序列、与近缘物种间序列变异的关系,并构建系统进化树。结果表明,火龙果叶绿体基因组133071 bp,呈环状双链四段式结构,包括大单拷贝区(LSC)68001 bp,小单拷贝区(SSC)21716 bp和两个12477 bp的反向重复区(IR)。火龙果叶绿体基因组共注释到基因118个,其中蛋白编码基因76个,rRNA基因4个、tRNA基因38个。火龙果叶绿体基因组密码子偏好以A/U(T)结尾,在检测到的66个简单重复序列(SSR)中,大多数是A/T碱基形成的单核苷酸重复基序。序列对比和系统进化分析显示,火龙果与同科的硫磺团扇和长枪团扇具有较近的亲缘关系。本研究首次测得火龙果叶绿体基因组全序列,明确其叶绿体基因组结构和序列特征,为火龙果的种植栽培和资源开发等研究提供了基因组信息。To illustrate the structure and sequence characteristics of pitaya(Selenicereus undatus)chloroplast genome and explore its phylogenetic relationship,the chloroplast genome of S.undatus was sequenced by high-throughput sequencing technology.Utilization of bioinformatics methods,the chloroplast genome sequence of pitaya was assembled and annotated,and its codon preference,simple sequence repeat,and sequence variation between related species were analyzed,a phylogenetic tree was also constructed.The results indicated that the full size of the chloroplast genome of pitaya was 133,071 bp,with a circular double-stranded four-segment structure,including 68,001 bp of large single copy region,21,716 bp of small single copy region,and two reverse repeat regions of 12,477 bp.A total of 118 genes were annotated in the chloroplast genome of pitaya,including 76 protein-coding genes,4 rRNA genes and 38 tRNA genes.The chloroplast genome of pitaya tended to end with A/U(T).Most of the detected 66 simple sequence repeats were the A/T bases formed mononucleotide repeats.Sequence comparison and phylogenetic analysis showed that S.undatus had a close relationship with Opuntia sulphurea and O.quimilo.The complete chloroplast genome of pitaya was detected for the first time in this study,and the structure and sequence characteristics of the chloroplast genome were clarified,which provided genomic information for the cultivation and resource development of pitaya.
