雪菊胎菊4种活性成分同时测定及其抗氧化活性研究

摘  要:目的:建立同时测定雪菊胎菊中4种活性成分含量的方法,并评估其体外抗氧化活性。方法:采用Kromasil 100-5-C_(18)(250 mm×4.6 mm,5μm)色谱柱分离,以甲醇(A)-0.1%磷酸水溶液(B)为流动相进行梯度洗脱,在348 nm测定雪菊胎菊中绿原酸、3,5-O-二咖啡酰基奎宁酸、槲皮苷及木犀草素的含量。分别采用DPPH法和ABTS法测定雪菊胎菊体外抗氧化活性。结果:在绿原酸线性范围为0.0350~0.02334 mg·mL^(-1)、3,5-O-二咖啡酰基奎宁酸线性范围为0.0347~0.2311 mg·mL^(-1)、槲皮苷线性范围为0.0269~0.1792 mg·mL^(-1)、木犀草素线性范围为0.0075~0.0500 mg·mL^(-1)下,R值均为0.9998,说明线性关系良好;平均加标回收率为98.10%~103.01%,相对标准偏差为0.25%~2.68%,说明该方法有较好的准确度。雪菊胎菊清除DPPH自由基的IC_(50)为2.4054 mg·mL^(-1),ABTS自由基的IC50为3.3772 mg·mL^(-1)。结论:该方法能高效且精确地同时测定4种活性成分,为雪菊胎菊的质量评估提供科学依据,且雪菊胎菊在抗氧化性能方面与雪菊全菊相当。Objective:Establish a method for simultaneous determination of four indicator components in Chrysanthemum buds and evaluate its in vitro antioxidant activity.Method:The Kromasil 100-5-C18(250 mm×4.6 mm,5μm)chromatographic column was used for separation,and methanol(A)-0.1%phosphoric acid aqueous solution(B)was used as the mobile phase.The contents of chlorogenic acid,3,5-O-dicaffeoylquinic acid,quercitrin,and luteolin in Chrysanthemum buds were determined at 348 nm.The in vitro antioxidant activity of Chrysanthemum buds was determined using DPPH and ABTS methods,respectively.Result:Under the linear ranges of 0.0350~0.02334 mg·mL^(-1) for chlorogenic acid,0.0347~0.2311 mg·mL^(-1) for 3,5-O-dicaffeoylquinic acid,0.0269~0.1792 mg·mL^(-1) for quercetin,and 0.0075~0.0500 mg·mL^(-1) for luteolin,the R values were 0.9998,indicating a good linear relationship.The average spiked recovery rate is 98.10% to 103.01%,and the relative standard deviation is 0.25%to 2.68%,indicating that this method has good accuracy.The IC50 values of DPPH radical scavenging rates for Chrysanthemum buds are 2.4054 mg·mL^(-1),the IC50 values of ABTS radical scavenging rates are 3.3772 mg·mL^(-1).Conclusion:This method can efficiently and accurately determine four active ingredients simultaneously,providing a scientific basis for the quality evaluation of Chrysanthemum buds.Chrysanthemum buds is comparable to Kunlun Chrysanthemum in terms of antioxidant performance.
