摘  要:目的探讨三阴型乳腺癌(triple-negative breast cancer,TNBC)中VISTA及Foxp3表达的临床病理意义。方法应用GEPIA2基于生物信息数据库TCGA分析VISTA mRNA、Foxp3 mRNA在TNBC中的表达量及与患者预后的关系。收集160例TNBC及160例正常乳腺组织样本,采用免疫组化MaxVision 3HRP法检测VISTA在肿瘤细胞和免疫细胞、Foxp3在肿瘤浸润淋巴细胞(tumor-infiltrating lymphocytes,TILs)中的表达及差异,并分析其与TNBC临床病理特征、预后的关系。结果数据库分析结果显示,VISTA mRNA在TNBC中表达水平低于正常乳腺组织(P<0.05),Foxp3 mRNA在TNBC中的表达水平高于正常乳腺组织(P<0.05);VISTA mRNA、Foxp3 mRNA在乳腺癌组织中表达呈正相关(r s=0.45,P<0.001);VISTA mRNA表达与患者总生存期有关。免疫组化结果显示,在TNBC中,肿瘤细胞VISTA阳性率为15%(24/160),免疫细胞VISTA阳性率为72.5%(116/160);Foxp3+TILs浸润密度高于正常乳腺组织(P<0.001);免疫细胞VISTA表达与Foxp3+TILs存在一定正相关性(r s=0.244,P=0.002);免疫细胞VISTA表达与患者TNM分期有关,Foxp3+TILs浸润密度与组织学分级有关;Cox单因素和多因素分析显示,TNM分期是影响TNBC患者预后的独立危险因素。结论在TNBC中,VISTA主要在免疫细胞表达,Foxp3+TILs浸润密度增高,免疫细胞VISTA表达及Foxp3+TILs高浸润密度与预后不良的临床病理特征相关,两者在TNBC不良进展中可能起协同作用。Purpose To investigate the clinicopathologic significance of VISTA and Foxp3 expression in triple-negative breast cancer(TNBC).Methods The expression of VISTA mRNA,Foxp3 mRNA in TNBC,along with their correlations with patient prognosis,were analyzed using GEPIA2 based on TCGA bioinformatics database.Samples of 160 TNBC tissues and 160 normal breast tissues were collected.Immunohistochemical detection was proformed using the MaxVision 3HRP method to detect the expression of VISTA in both tumor cells and immune cells,and Foxp3 in tumor-infiltrating lymphocytes(TILs).Their differential expression,as well as the relationships with the clinicopathological characteristics and prognosis in TNBC,were analyzed.Results Database analysis results showed that the expression level of VISTA mRNA in TNBC was lower than that in normal breast tissue(P<0.05),while the expression level of Foxp3 mRNA in TNBC was higher than that in normal breast tissue(P<0.05).The expression of VISTA mRNA and Foxp3 mRNA in breast cancer tissues were positively correlated(r s=0.45,P<0.001),and the expression of VISTA mRNA was related to the overall survival.Immunohistochemical results showed that in TNBC,the positive expression rate of VISTA in tumor cells was 15%(24/160),and 72.5%(116/160)in immune cells.The infiltration density of Foxp3+TILs was higher than that in normal breast tissue(P<0.001).The expression of VISTA in immune cells was positively correlated with Foxp3+TILs(r s=0.244,P=0.002).Moreover,the expression of VISTA in immune cells was related to the TNM stage of patients,and the infiltration density of Foxp3+TILs was related to the histological grade.Cox univariate and multivariate analyses indicated that TNM stage was an independent risk factor affecting the prognosis of TNBC patients.Conclusion In TNBC,VISTA is mainly expressed in immune cells and an increased infiltration density of Foxp3+TILs is observed.Both VISTA expression in immune cells and high infiltration density of Foxp3+TILs are correlated with clinicopathologic features ind
