摘  要:目的 筛选与膀胱癌免疫检查点抑制剂(ICIs)治疗效果相关的关键基因。方法利用生物信息学方法,获取IMvigor 210数据集中同时与ICIs治疗效果及总生存率(OS)相关的基因,利用这些基因将癌症基因组图谱(TCGA)数据库中膀胱癌患者进行亚组分型,通过对不同亚组的差异表达基因进行蛋白相关(PPI)网络构建筛选关键基因。结果 共识别出与OS相关的基因1 455个(均P<0.05),免疫治疗反应组与无反应组差异表达基因为91个(倍数>1.5倍,均P<0.05),二者之间交集为31个基因。TCGA数据库中膀胱癌患者共分为两个亚组,两亚组间生存率差异有统计学意义(P=0.016),中位生存时间分别为1.8年和3.8年,HR(95%CI):[1.449,(1.070~1.962)]。两亚组共识别差异表达基因1 346个(倍数>2,均P<0.05),差异表达基因对应蛋白进行PPI网络构建中超过5个相互作用关系的蛋白共12个,分别为EGFR、SRC、ERBB2、HLA-DRA、B2M、C1QA、MT-CYB、MT-ND1、MT-ND2、MT-ND4L、MT-ND5和MT-ND6。结论筛选出的12个关键基因具有预测膀胱癌患者对ICIs治疗反应的潜力。Objective To screen key genes related to the efficacy of immune checkpoint inhibitors(ICIs)in bladder cancer.Methods Bioinformatics were used to obtain genes related to ICIs treatment efficacy and overall survival(OS)in the IMvigor 210 dataset.These genes were used to subtype bladder cancer patients in the TCGA database.Protein-protein interaction(PPI)network construction was used to screen key genes from the differentially expressed genes between the two subgroups.Results A total of 1455 genes related to OS(P<0.05)and 91 differentially expressed genes between the ICIs response group and the non-response group(fold change>1.5,P<0.05)were identified,and the intersection of the two was 31 genes.Two subgroups of bladder cancer patients in the TCGA database were identified,and the survival rate between the two subgroups was statistically different(P=0.016),with a median survival time of 1.8 years compared to 3.8 years,HR(95%CI):[1.449,(1.070~1.962)].A total of 1346 differentially expressed genes(fold change>2,P<0.05)were identified between the two subgroups,and 12 proteins with more than 5 mutual interaction relationships were identified in the PPI network construction of the corresponding proteins,including EGFR,SRC,ERBB2,HLA-DRA,B2M,C1QA,MT-CYB,MTND1,MT-ND2,MT-ND4L,MT-ND5 and MT-ND6.Conclusions The 12 key genes have the potential to predict the response of bladder cancer patients to ICIs treatment.
